Carbapenem-resistant Acinetobacter baumannii is recognized by the World Health Organization (WHO) as one of top priority pathogens. Despite its public health importance, genomic data clinical isolates from Malaysia remain scarce. In this study, whole-genome sequencing was performed on 126 A . collected main tertiary hospital in state Terengganu, Malaysia, over a 10-year period (2011–2020). Antimicrobial susceptibilities determined for 20 antibiotics belonging to 8 classes showed that 77.0% ( n =97/126) were categorized multidrug resistant (MDR), with all MDR being carbapenem resistant. ABSTRACT Acinetobacter baumannii is highly resistant to antimicrobial agents, and XDR strains have become widespread. A. has developed resistance colistin, which considered the last resort against Gram-negative bacteria, mainly caused by lipooligosaccharide (LOS) phosphoethanolamine (pEtN) and/or galactosamine (GalN) modifications induced mutations that activate two-component system (TCS) pmrAB . The genus Acinetobacter comprises both environmental and clinically relevant species associated with hospital-acquired infections. Among them, baumannii is a critical priority bacterial pathogen, for which the research development of new strategies antimicrobial treatment are urgently needed. spp. produce variety structurally diverse capsular polysaccharides (CPSs), surround cells thick protective layer. These surface structures primary receptors capsule-specific bacteriophages, that is, phages carrying tailspikes CPS-depolymerizing/modifying activities. Phage tailspike proteins (TSPs) exhibit hydrolase, lyase, or esterase activities toward corresponding CPSs certain structure. In this study, data on all lytic infecting genomes deposited in NCBI GenBank database by January 2024 were summarized. 149 identified TSPs encoded 143 phages, specificity (K specificity) 46 has been experimentally determined predicted previously. 63 CPSs, produced various K types, was study using bioinformatic analysis. A comprehensive phylogenetic analysis confirmed prediction revealed possibility genetic exchange gene regions to CPS-recognizing/degrading parts different between morphologically taxonomically distant groups phages.

Abstract Many bacterial pathogens, including the human exclusive pathogen Salmonella Typhi, express capsular polysaccharides as a crucial virulence factor. Here, through S . Typhi whole genome sequence analyses and functional studies, we found list of single point mutations that make hypervirulent. We discovered mutation in Vi biosynthesis enzymes control polymerization or acetylation is enough to result different capsule variants Typhi. All variant strains are pathogenic, but hyper particularly hypervirulent, demonstrated by high morbidity mortality rates observed infected mice. The hypo have primarily been identified Africa, whereas distributed worldwide. Collectively, these studies increase awareness about existence establish solid foundation for numerous future on variants, offer valuable insights into strategies combat capsulated bacteria.

Acinetobacter baumannii and Klebsiella pneumoniae are significant nosocomial pathogens worldwide. In this study, the general characterization of A. K. isolates obtained from blood intensive care unit patients multidisciplinary scientific practical center emergency medicine January to September 2024 was performed. Prophage regions prophage-derived tailspike polysaccharide-depolymerizing or -modifying enzymes within these were identified characterized in detail using a refined workflow. The protocol, encompassing comprehensive survey all predicted bacterial proteins, revealed an average 6.0 prophage per genome, including putatively derived filamentous phages, 4.8 isolate. Analysis putative indicated that most related previously isolated, yet unclassified, temperate phages infecting pneumoniae. However, certain sequences likely originated representing novel groups comparatively distant known phages.
