Abstract Background Achieving complete immune reconstitution (CIR) in people with human immunodeficiency virus (PWH) following antiretroviral therapy (ART) is essential for preventing acquired syndrome (AIDS) progression and improving survival. However, there a paucity of robust prediction models determining the likelihood CIR PWH after ART. We aimed to develop validate model utilizing baseline data. Methods Data including demographic information, immunological profiles, routine laboratory test results, were collected from Yunnan, China. The participants divided into training validation sets (7:3 ratio). To construct accompanying nomogram, univariate multivariate Cox regression analyses performed. was evaluated using C-index, time-dependent receiver operating characteristic (ROC) curves, calibration clinical decision curves assess discrimination, calibration, applicability. Results 5 408 included, 38.52%. analysis revealed various independent factors associated CIR, infection route, marital status, CD4+ T cell count, CD4/CD8 ratio. A nomogram formulated predict probability achieving at years 4, 5, 6. demonstrated good performance, as evidenced by an AUC 0.8 both sets. Amphetamine-type stimulant (ATS) abuse is strongly associated with an elevated risk of HIV infection and transmission. Antiretroviral therapy (ART) serves as the primary approach for managing AIDS progression. However, ATS diminishes efficacy ART in HIV/AIDS patients, amplifying vulnerability to immunological non-response (INR) ultimately increasing incidence rate mortality opportunistic infections. Currently, no effective interventions targeting INR exist. Acupuncture has demonstrated promise bidirectionally modulating body's immune response may be beneficial combined abuse. Nevertheless, further research comprehensive evaluation are imperative substantiate these findings. This study a two-center, randomized, non-acupoint controlled, single-blind clinical trial. It will conducted two large drug rehabilitation centers western China, involving 114 patients receiving ART. The participants randomly assigned either Transcutaneous Electrical Acupoint Stimulation (TEAS) + group or sham-TEAS group, 1:1 ratio. Both groups receive 48-week treatment. outcome measure assessed after treatment CD4 T cell count. Secondary measures include reconstitution efficiency CD4/CD8 ratio, CD45RA CD45RO counts, CD28 CD38 CD8 ki67 JC mitochondrial membrane potential testing, infections, PRO scale.
