Describing the microbial community within tumour has been a key aspect in understanding pathophysiology of microenvironment. In head and neck cancer (HNC), most studies on tissue samples have only performed 16S rRNA short-read sequencing (SRS) V3-V5 region. SRS is mostly limited to genus level identification. this study, we compared full-length long-read (FL-ONT) from Oxford Nanopore Technology (ONT) V3-V4 Illumina (V3V4-Illumina) 26 HNC tissues. Further validation was also using culture-based methods 16 bacterial isolates obtained 4 patients MALDI-TOF MS. We observed similar alpha diversity indexes between FL-ONT V3V4-Illumina. However, beta-diversity significantly different techniques (PERMANOVA - R

A growing body of research indicates that a wide range cancer types may correlate with human microbiome components. On the other hand, little is known about potential contribution oral microbiota to cancer. However, some components can stimulate different tumorigenic processes associated development In this line, two prevalent infections, Porphyromonas gingivalis, and Fusobacterium nucleatum increase tumor growth. The impact course illness through direct interactions major modifications toxicity responsiveness kinds therapy. Recent has demonstrated relationship between specific phylogenetic groupings results immunotherapy treatment for particular types. Conversely, there been recent upsurge in interest possibility using microbes treat At moment, species, such as Salmonella typhimurium Clostridium spp., are being explored possible vectors. Thus, understanding these microbial highlights importance maintaining healthy preventing cancers. From perspective, review will discuss role on cancers their application treatment/improvement.

The human microbiota comprises a diverse microbial ecosystem that significantly impacts health and disease. Among its components, the gut plays crucial role in regulating metabolic, immunologic, inflammatory responses. Dysbiosis, an imbalance composition, has been linked to carcinogenesis through mechanisms such as chronic inflammation, metabolic disturbances, epigenetic modifications, immune system dysregulation. Additionally, dysbiosis influences efficacy toxicity of cancer therapies. Given these associations, there is growing interest leveraging biomarker for detection outcome prediction. Notably, distinct signatures have identified across various types, suggesting their potential diagnostic markers. Furthermore, modulation presents promising avenue improving treatment outcomes strategies antibiotics, prebiotics, probiotics, fecal transplantation, dietary interventions, small-molecule inhibitors, phage therapy. To explore relationships, we conducted comprehensive literature review using Web Science, Scopus, PubMed, MEDLINE, Embase, Google Scholar our primary online databases, focusing on indexed peer-reviewed articles up present year. This aims elucidate development, examine molecular involved, assess impact highlight microbiota-based therapeutic discuss applications management. A deeper understanding intricate interplay between may pave way novel approaches prevention, early detection, optimization.

Oral squamous cell carcinoma (OSCC) is a highly aggressive malignancy, with low 5-y survival rate and frequent local recurrence or metastasis. This study explores the role of γδT cells in development progression OSCC. cells, which exhibit innate adaptive immune characteristics, are known for their dual cancer, acting as anti- protumor agents depending on context. Using murine model OSCC induced by carcinogen 4-nitroquinoline-1-oxide (4NQO), adequately mimics human OSCC, we investigated impact tumor growth microenvironment. We first characterized tongue epithelium, primary site cancer this model. The results indicate that predominantly Vγ6+ subset, expanding postnatally microbiota-dependent manner. Upon 4NQO administration, depletion did not significantly alter kinetics but result reduction size number, suggesting promoting growth. Interestingly, absence IL-17, key cytokine produced also resulted reduced volume without affecting disease progression, corroborating these Further analysis revealed IL-17-producing facilitate angiogenesis within microenvironment expression angiogenic factors. Of note, while treatment increased oral microbial load altered its composition, IL-17 deficiency affect microbiota, indicating effects independent changes. highlights pathologic particularly through angiogenesis. underscores importance need further research into therapeutic strategies targeting cells.

Oral cavity dysbiosis is associated with numerous inflammatory diseases, including diabetes, bowel and periodontal disease. Changes in the oral microenvironment lead to bidirectional interactions between pathogens individual host systems, which may induce systemic inflammation. Digestive system tumours (DSTs) often diagnosed late due to nonspecific symptoms. Non-invasive biomarkers are crucial for early detection and improved outcomes. We collected tongue coating samples from 710 patients with DST 489 healthy controls (HC) April 2023, December 2023. Microbial composition was analyzed using 16S rRNA sequencing, five machine learning algorithms were applied assess the diagnostic potential of microbiota. Alpha diversity analysis showed that microbial in significantly increased patients. LEfSe identified DST-enriched genera Alloprevotella Prevotella, contrasting HC-dominant taxa Neisseria, Haemophilus, Porphyromonas (LDA >4). Notably, when comparing each four subtypes HC group, proportion Haemophilus group higher, it as an important feature distinguishing group. Machine validation demonstrated superior performance Extreme Gradient Boosting (XGBoost) model, achieving AUC 0.926 (95% CI: 0.893-0.958) internal validation, outperforming other models.
