Bacillus cereus translocator protein (BcTSPO) is a transmembrane that plays regulatory role in various phenotypes, potentially including its virulence. Given the exoproteome major determinant of B. virulence, particularly during late stages active growth, analysis crucial for understanding functions BcTSPO. In this study, we conducted comparative exoproteomes and cellular proteomes Δtspo mutant wild-type ATCC 14579 strain exponential early stationary growth phases. Shotgun proteomics revealed absence BcTSPO significantly depleted secreted virulence factors, HBL, NHE, CytK enterotoxins. Interestingly, cholesterol-dependent cereolysin O (CLO) was more abundant exoproteome, indicating distinct mechanism. The proteome further confirmed positively regulates, likely indirectly, synthesis enterotoxins negatively regulates production CLO. Additionally, enriched moonlighting proteins, reflecting impact on metabolism stress responses. Overall, these findings highlight critical shaping composition both proteome, with significant implications cereus.

Mycoplasma bovis is the most common mycoplasma associated with cattle diseases worldwide. However, other seemingly less virulent spp. such as M. bovigenitalium and bovirhinis have also been mycoplasmosis. The study objective was to compare adhesion cellular invasion characteristics of these bovine using Madin–Darby Bovine Kidney (MDBK) epithelial cells. MDBK cells were separately infected 12 strains one strain each bovirhinis. Following infection, a gentamicin protection assay performed lysed at 6 54 h post-infection. cell lysates cultured for qPCR used estimate average number bacterial that (Myc/Cell ratio). Confocal electron microscopy studies mNeonGreen performed. All 14 multiplied within cells, finding confirmed by strain. Unexpectedly, strains, obtained from diseased asymptomatic bison, had lower Myc/Cell ratios than strains. These findings suggest ability mycoplasmas invade replicate host does not account differences in virulence between species.
