The incidence of cervical cancer caused by human papillomavirus (HPV) infection has increased in recent years. More than half all cases are due to HPV16 and HPV18 infection, so testing is essential prevent cancer. HPV mainly carried out hospitals, but it subject time specialized medical facilities. On the other hand, home self-testing using simple diagnostics would present an attractive alternative privacy flexibility with regard place, provided sufficient sensitivity specificity can be achieved. In this work, a dual lateral flow assay based on RPA-CRISPR-Cas12a/13a (named RC-LFA) for detection was described. Taking advantage cleavage Cas12a Cas13a, CRISPR-Cas12a/Cas13a system designed detect HPV18. strip two test lines suit CRISPR-Cas12a/Cas13 system. RC-LFA achieves rapid simultaneous high (10 copies/μL) about 40 min from extraction nucleic acid instrument-free readout. user-friendly instrument-free, making promising method self-tests at home.

Abstract Nucleic acid detection plays a crucial role in various applications, including disease diagnostics, research development, food safety, and environmental health monitoring. A rapid, point‐of‐care (POC) nucleic test can greatly benefit healthcare system by providing timely diagnosis for effective treatment patient management, as well supporting diseases surveillance emerging pandemic diseases. Recent advancements acids technology have led to rapid assays single‐stranded that be integrated into simple miniaturized platforms ease of use. In this review, the study focuses on developments isothermal amplification, hybridization circuits, enzyme‐based signal reporting mechanisms, show promise POC testing. The also evaluates critical technical breakthroughs identify advantages disadvantages these methods applications.

Diagnostic testing plays a key role in surveillance program as diagnostic aims to accurately determine the infection or disease status of an individual animal. assays for AIV can be categorized into four broad types: tests detecting virus, its antigen, genomic material, and antibodies virus. Virus characterization almost always follows virus detection. The present article surveys current literature on goals, principles, test performance, advantages, disadvantages these assays. isolation achieved using embryonating eggs cell cultures lab setting. antigens detected by antigen-capturing immunoassays tissue immunoassays. ABSTRACT Aflatoxin contamination in food and feed is a global concern for safety, posing great threat to human environmental health. It the secondary metabolite mainly produced by Aspergillus flavus ( A. ) parasiticus (A. ). Therefore, it very urgent develop rapid sensitive identification detection methods aflatoxigenic fungi realize early warning risk of aflatoxin from source. This article reviews latest research progress strategy identifying detecting agricultural recent years. The principles applications different techniques determination, including morphological identification, nucleic acid amplification techniques, spectral analysis technology, biosensing, are presented this review. Finally, challenges trends future also discussed. Through fungi, there will be relatively sufficient time do corresponding prevention control measures reduce or even prevent production stage, which beneficial greatly improve safety.

Members of the Pol A family DNA polymerases, found across all domains life, utilize various strategies for strand separation during replication. In higher eukaryotes, mitochondrial polymerase γ relies on replicative helicase TWINKLE, whereas yeast ortholog, Mip1, can unwind independently. Using Mip1 as a model, we present series high-resolution cryo-EM structures that capture process displacement. Our data reveal previously unidentified structural elements facilitate unwinding downstream duplex. Yeast cells harboring variants defective in displacement exhibit impaired oxidative phosphorylation and loss mtDNA, corroborating observations. This study provides molecular basis intrinsic activity illuminates distinct mechanisms utilized by polymerases. Background. Hypothermic machine perfusion (HMP) has greatly improved kidney allograft preservation. However, tissue damage still occurs during HMP, affecting posttransplant graft function. Therefore, better methods are needed to continuously assess organ quality and predict function survival. We propose that soluble DNA (sDNA) concentration in HMP perfusate can be used as a noninvasive biomarker for this purpose. Methods. Perfusate samples of grafts placed on were collected 5 min after the initiation again at conclusion HMP. sDNA nuclear origin from was quantified by real-time polymerase chain reaction correlated with parameters clinical outcomes. Addressing the pervasive issue of food adulteration and fraud driven by economic interests has long presented a complex challenge. Such not only compromises safety supply chain destabilizes market economy but also poses significant risks to public health. Food encompasses practices such as substitution, process manipulation, mislabeling, introduction undeclared ingredients, genetically modified foods. Given diverse range deceptive methods employed, genomics-based identification techniques have increasingly been utilized for detecting adulteration. Compared traditional detection methods, technologies polymerase reaction (PCR), next-generation sequencing (NGS), high-resolution melt (HRM) analysis, DNA barcoding, CRISPR-Cas system demonstrated efficacy in accurately sensitively even trace amounts adulterants. This paper provides an overview approaches identifying adulteration, summarizes latest applications certification procedures, discusses current limitations, explores potential future trends, thereby offering new insights enhance control quality contributing development more robust regulatory frameworks policies.
