Paeonol, as one of the most abundant plant-derived polyphenols, has multiple bioactivities including anti-inflammatory, anti-tumor, and anti-cardiovascular diseases. Nevertheless, anti-aging effects related mechanisms paeonol are rarely reported. In this study, we found that significantly prolonged mean lifespan Caenorhabditis elegans (C. elegans) by 28.49% at a dose 200 μM. Moreover, promoted health C. increasing body bending pharyngeal pumping rates reducing lipofuscin accumulation level. Meanwhile, induced expression stress-related genes or proteins activating transcription factors DAF-16/FOXO, SKN-1/Nrf2, HSF-1, which in turn enhanced oxidative thermal stress tolerance. The mechanism behind effect occurred down-regulating insulin/IGF-1 signaling (IIS) pathway. Our findings shed new light on application for longevity promotion human health.

Aminoglycoside antibiotic-induced sensorineural hearing loss (SNHL) is a common sensory disorder that requires the development of prophylactic and therapeutic interventions. Lycium barbarum glycopeptide (LBGP) peptidoglycan isolated purified from polysaccharides exhibit significant anti-inflammatory, antioxidant, neuroprotective effects, but role LBGP in aminoglycoside-induced SNHL has not been well investigated. Here it shown can protect against neomycin-induced impairment alleviate oxidative stress mouse model. Moreover, further found inhibition tryptophan hydroxylase (Tph)-mediated serotonin (5-HT) biosynthesis plays key mechanism action treating loss. Systemic delivery 5-HT increased apoptosis cochlear hair cells spiral ganglion neurons, pharmacological Tph2 with P-chlorophenylalanine or knock down by AAV-ie-Tph2 effectively attenuated dysfunction. Collectively, these results provide promising strategy for prevention using natural plant extract which more available exhibits lower side effects compared other otoprotective drugs, identify as potential target treatment ototoxicity.
