Rodents, in particular rats and mice, are currently the most widely employed animal models psychology behavioral neuroscience. Nevertheless, an interesting historical question is: when was first rodent study performed by whom? The current article presents history of science: a case interspecies social bonding between rat dog, observed 1822 British chemist Samuel Moss (1794–1868) subsequently described same scientific 1836. In present article, after biographical sketch Moss, I examine detail notable Moss. This is under several points view. First, Moss’s albino, variety which at that time extremely rare. Existing models of frontotemporal dementia (FTD) may not fully recapitulate the pathophysiology disease. To generate more pathophysiologically relevant FTD models, we engineered MAPT knockin mouse lines carrying triple mutations, among which MAPTP301S;Int10+3;S320F line exhibited robust tau pathology starting before 6 months age. Severe accumulation was predominantly observed in thalamus, hypothalamus, and amygdala with milder involvement cortex hippocampus, leading to synaptic loss, brain atrophy, FTD-like behavioral abnormalities. Crossbreeding mice App knockin, AppNL-G-F, markedly enhanced highlighting interplay between β-amyloid tau. These findings establish mutant as valuable for investigating mechanisms underlying other tauopathies, providing a platform vivo drug screening.

Abstract We recently reported development of human MAPT knock-in mice that carry single or double pathogenic mutations frontotemporal dementia. However, it takes more than 14 months for the line with most aggressive phenotypes to exhibit tau pathology without forming high-order oligomers, along concomitant abnormal behavior. thus generated carrying triple mutations, among which P301S;Int10+3;S320F exhibited robust starting earlier 6 months. Tau accumulation took place mainly in thalamus, hypothalamus, amygdala and entorhinal cortex, but less so hippocampus, leading synaptic loss, atrophy behavioral abnormalities. Crossbreeding App NL-G-F resulted manifestation hippocampus cortex. These mutant will be valuable tools understanding mechanisms dementia, Alzheimer’s disease other tauopathies.
