Abstract Messenger RNA (mRNA) vaccines against COVID‐19 are the first authorized biological preparations developed using this platform. During pandemic, their administration has been proven to be a life‐saving intervention. Here, we review main advantages of mRNA vaccines, identify further technological challenges met during development platform, and provide an update on clinical progress leading vaccine candidates different viruses that include influenza viruses, human immunodeficiency virus 1, respiratory syncytial virus, Nipah Zika cytomegalovirus, Epstein‐Barr virus. The prospects manufacturing in low‐income countries also discussed. ongoing interest research technology likely overcome some existing for (e.g., related storage conditions immunogenicity components lipid nanoparticles) enhance portfolio diseases which classical formulations already authorized. Neurofilament light chain (NF-L) concentration is recognized to be modified in neurological diseases and traumatic brain injuries, but studies the normal aging population are lacking. It is, therefore, urgent identify influencing factors of NF-L population. We assessed sera a large cohort 409 community-dwelling adults aged over 65 years. We studied association between various physiological also with self-reported comorbidities or life-style habits. We showed that serum was tightly associated cystatin C (r = 0.501, p < 0.0001) consequently, estimated glomerular filtration rate (eGFR) - 0.492; 0.0001). Additionally, dependent on age body mass index (BMI) not sex. Among comorbidities, subjects who reported disorders, cardiovascular history fracture had higher univariate analysis, whereas it only case for disorders multivariate analysis. increased when Mini-Mental State Examination (MMSE) decreased (≤ 25 points) geriatric depression score (GDS) (> 5 both Finally, we providing reference ranges by categories without altered renal function.NF-L driven increasing number depression. Abstract Aims/hypothesis No established blood-based biomarker exists to monitor diabetic sensorimotor polyneuropathy (DSPN) and evaluate treatment response. The neurofilament light chain (NFL), a blood of neuroaxonal damage in several neurodegenerative diseases, represents potential for DSPN. We hypothesised that higher serum NFL levels are associated with prevalent DSPN nerve dysfunction individuals recently diagnosed diabetes. Methods This cross-sectional study included 423 adults type 1 2 diabetes known duration less than year from the prospective observational German Diabetes Study cohort. was measured samples fasting participants multiplex approach using proximity extension assay technology. assessed by neurological examination, conduction studies quantitative sensory testing. Associations (defined according Toronto Consensus criteria) were estimated Poisson regression, while multivariable linear quantile regression models used assess associations function measures. In exploratory analyses, other biomarkers panel also analysed similarly NFL. Results found 16% sample. Serum increased age. After adjustment age, sex, waist circumference, height, HbA 1c , duration, type, cholesterol, eGFR, hypertension, CVD, use lipid-lowering drugs non-steroidal anti-inflammatory drugs, (RR [95% CI] per 1-normalised protein expression increase, 1.92 [1.50, 2.45], p <0.0001), slower motor (all <0.0001) ≤0.03) velocities, lower sural action ( =0.0004) thermal detection threshold warm stimuli =0.023 =0.004 hand foot, respectively). Serum neurofilament light chain (sNfL) levels correlate with multiple sclerosis (MS) disease activity, but the dynamics of this correlation are unknown. We evaluated relationship between sNfL and radiologic MS activity through monthly assessments during 24-week natalizumab treatment interruption period in RESTORE (NCT01071083).

Background Neurofilament light chain (NfL) in cerebrospinal fluid (CSF) is a biomarker of multiple sclerosis (MS). However, CSF sampling invasive and has limited the clinical application. With development highly sensitive single-molecule assay, accurate quantification very low NfL levels blood become feasible. As evidence being accumulated, we performed meta-analysis to evaluate diagnostic predictive value MS patients. Methods We literature search on PubMed, EMBASE, Web Science Cochrane Library from inception May 31, 2022. The differences between vs . controls, clinically isolated syndrome (CIS), progressive (PMS) relapsing-remitting (RRMS), relapse remission were estimated by standard mean difference (SMD) corresponding 95% confidence interval (CI). Pooled hazard ratio (HR) 95%CI calculated predict time reach Expanded Disability Status Scale (EDSS) score≥4.0 relapse. Results A total 28 studies comprising 6545 patients 2477 controls eligible for diagnosis value, 5 with 4444 synthesized analysis value. Blood significantly higher age-matched (SMD = 0.64, 0.44–0.85, P<0.001), vs. non-matched 0.76, 0.56–0.96, P<0.001) CIS 0.30, 0.18–0.42, PMS RRMS 0.56, 0.27–0.85, relapsed remitted 0.54, 0.16–0.92, P 0.005). Patients high had shorter EDSS (HR 2.36, 1.32–4.21, 0.004) but similar 1.32, 0.90–1.93, 0.155) compared those levels. Conclusion far as know, this first evaluating MS. present study indicates may be useful diagnosing MS, distinguishing subtypes predicting disease worsening future.
