Abstract Messenger RNA (mRNA) vaccines against COVID‐19 are the first authorized biological preparations developed using this platform. During pandemic, their administration has been proven to be a life‐saving intervention. Here, we review main advantages of mRNA vaccines, identify further technological challenges met during development platform, and provide an update on clinical progress leading vaccine candidates different viruses that include influenza viruses, human immunodeficiency virus 1, respiratory syncytial virus, Nipah Zika cytomegalovirus, Epstein‐Barr virus. Abstract Aims/hypothesis No established blood-based biomarker exists to monitor diabetic sensorimotor polyneuropathy (DSPN) and evaluate treatment response. The neurofilament light chain (NFL), a blood of neuroaxonal damage in several neurodegenerative diseases, represents potential for DSPN. We hypothesised that higher serum NFL levels are associated with prevalent DSPN nerve dysfunction individuals recently diagnosed diabetes. Methods This cross-sectional study included 423 adults type 1 2 diabetes known duration less than year from the prospective observational German Diabetes Study cohort. was measured samples fasting participants multiplex approach using proximity extension assay technology. assessed by neurological examination, conduction studies quantitative sensory testing. Associations (defined according Toronto Consensus criteria) were estimated Poisson regression, while multivariable linear quantile regression models used assess associations function measures. Background and Objectives

To investigate whether white matter lesion activity, acute axonal damage, density in MS associate with CSF neurofilament light chain (NfL) levels.

Methods

Of 101 brain donors (n = 92 progressive MS, n 9 relapsing-remitting MS), ventricular was collected, NfL levels were measured. White lesions classified as active, mixed, inactive, or remyelinated, microglia/macrophage morphology active mixed ramified, ameboid, foamy. In addition, damage assessed using Bielschowsky amyloid precursor protein (APP) (immune)histochemistry.

Results

measurements of recent (<1 year) clinically silent stroke excluded. correlated negatively disease duration (p = 6.9e-3, r 0.31). without atrophy, positively the proportion containing foamy microglia/macrophages 9.85e-10 p 1.75e-3, respectively), but not those ramified microglia. proportions inactive 5.66e-3) remyelinated 0.03). normal appearing pyramid tract, (Bielschowsky, 0.02, −0.31), presence related to higher (APP, 1.17e-6). The amount rim when compared 4.6e-3 center border (center: 4.6e-3, border, n.s., p 0.03 0.04, respectively).

Discussion

Our results demonstrated that microglia show high correlate elevated data support use this biomarker monitor inflammatory demyelinating activity MS.

Background Neurofilament light chain (NfL) in cerebrospinal fluid (CSF) is a biomarker of multiple sclerosis (MS). However, CSF sampling invasive and has limited the clinical application. With development highly sensitive single-molecule assay, accurate quantification very low NfL levels blood become feasible. As evidence being accumulated, we performed meta-analysis to evaluate diagnostic predictive value MS patients. Methods We literature search on PubMed, EMBASE, Web Science Cochrane Library from inception May 31, 2022. The differences between vs . controls, clinically isolated syndrome (CIS), progressive (PMS) relapsing-remitting (RRMS), relapse remission were estimated by standard mean difference (SMD) corresponding 95% confidence interval (CI). Pooled hazard ratio (HR) 95%CI calculated predict time reach Expanded Disability Status Scale (EDSS) score≥4.0 relapse. Results A total 28 studies comprising 6545 patients 2477 controls eligible for diagnosis value, 5 with 4444 synthesized analysis value. Blood significantly higher age-matched (SMD = 0.64, 0.44–0.85, P<0.001), vs. non-matched 0.76, 0.56–0.96, P<0.001) CIS 0.30, 0.18–0.42, PMS RRMS 0.56, 0.27–0.85, relapsed remitted 0.54, 0.16–0.92, P 0.005). Patients high had shorter EDSS (HR 2.36, 1.32–4.21, 0.004) but similar 1.32, 0.90–1.93, 0.155) compared those levels. Conclusion far as know, this first evaluating MS. present study indicates may be useful diagnosing MS, distinguishing subtypes predicting disease worsening future.
