Multiple sclerosis (MS) is a complex inflammatory disease affecting the central nervous system. Most commonly, it begins with recurrent symptoms followed by partial or complete recovery, known as relapsing–remitting MS (RRMS). Over time, many RRMS patients progress to secondary progressive (SPMS), marked gradual symptom deterioration. The factors triggering this transition remain unknown, lacking predictive biomarkers. This study aims identify blood biomarkers specific SPMS. We analyzed six datasets of SPMS and patients’ brain tissues, compared differential expressed genes (DEGs) obtained highlight DEGs reflecting alterations occurring in both tissues potential biological processes involved. observed total 38 up-regulated their interaction network was evaluated through analysis. Among aforementioned DEGs, 21 may be directly involved transition. Further, we highlighted three processes, including calcineurin–NFAT pathway, related investigated serve promising means monitor from SPMS, which still elusive. Given that they can also sourced samples, approach could offer relatively rapid convenient method for monitoring facilitating expedited assessments.
