Perineuronal nets (PNNs) are specialized extracellular matrix structures that predominantly surround inhibitory neurons in the central nervous system (CNS). They have been identified as crucial regulators of synaptic plasticity and neuronal excitability. This literature review aims to summarize current state knowledge about PNNs, their molecular composition structure, well functional roles involvement neurological diseases. Furthermore, future directions PNN research proposed, therapeutic potential targeting PNNs develop novel treatment options for various disorders is explored. emphasizes importance CNS physiology pathology underscores need further this area.

Abstract The CSF1R inhibitor Plx3397, an FDA-approved treatment for a rare cancer, has been shown to reduce microglia, lower inflammation, and improve synaptic integrity in several mouse models of Alzheimer’s disease (AD). However, the effects Plx3397 on neural function AD remain largely unknown. Here, we used 5xFAD model characterize effect treatment. While administration increased density mice, it also reduced percentage neurons phase-locked gamma oscillations. This decoupling was closely associated with gene expression changes related synapse organization. These observations prompted us investigate whether gamma-frequency entrainment could counteract circuit alterations induced by administration. We thus enhanced phase-locking using non-invasive patterned sensory light stimulation. Remarkably, restoring oscillations improved function, reshaped signatures, learning memory Plx3397-treated mice. findings suggest that inhibitors like may benefit from multimodal approach combining microglial targeting stimulation support physiology cognitive AD.
