An essential function of memory is to guide behavior for survival and adaptation. While considerable knowledge has been accumulated on formation, much less understood about how retrieved memories direct behavior/action. In the auditory Pavolovian threat conditioning paradigm, retrieval conditioned activates dorsomedial prefrontal (dmPFC) neurons exhibiting transient responses (T-neurons), which activate both dmPFC sustained (S-neurons) locus coeruleus (LC) neurons. Auditory inputs S-neurons enable conversion from so that freezing durations match those cues. Activation LC required by enhancing S-neuron responses, which, interestingly, opens a short time window during non-conditioned cues also lead freezing. The transition thus hinges integration memory, sensory inputs, emotional/body state generate selective, adequate, finely tuned behavior.

Abstract Background Maladaptive emotion regulation plays a crucial role in the development and maintenance of elevated anxiety levels, both patients individuals with subclinical symptomatology. While pharmacological treatments for target dysregulation through dopaminergic, noradrenergic serotonergic systems, little is known about underlying mechanisms. Therefore, current study depicts association these neuromodulatory systems’ resting-state functioning trait-anxiety, investigating maladaptive regulation. Methods Amplitude low-frequency fluctuations (ALFF), fractional amplitude (fALFF), whole-brain functional connectivity (rs-FC) were obtained from ventral tegmental area (VTA), locus coeruleus (LC) dorsal raphe, correlated trait-anxiety self-reported (N = 60). Results Trait-anxiety was positively associated LC’s fALFF negatively VTA’s rs-FC left inferior parietal lobule (L-IPL) superior frontal gyrus (L-SFG). regions, fully mediating this association. VTA region, but not parietal, predicted its neural oscillations, an effect that paralleled by stronger dopaminergic innervation. Limitations Use external molecular datasets need replication patients. Conclusions Noradrenergic systems appear to contribute differently anxiety. noradrenaline likely acts more general mechanism, dysconnectivity frontoparietal control network may act as one mechanisms regulation, informing models on disorder development.
