In
eukaryotes,
microRNAs
(miRNAs)
are
generated
by
Dicer/Dicer-Like
(DCL)-mediated
cleavage.
Previous
studies
identified
three
DCL
genes
(CrDCL1-3)
in
Chlamydomonas
reinhardtii
and
indicated
that
CrDCL3
mediated
the
production
of
most
miRNAs,
while
CrDCL1
protein
was
mainly
involved
siRNA
biogenesis.
The
role
miRNA
biogenesis
remains
unclear.
This
study
constructed
a
phylogenetic
tree,
performed
structural
analyses
Dicer/DCL
proteins
from
multiple
species
screened
verified
dcl1
dcl3
mutant
strains.
Using
CC-5325
as
control
groups,
we
sRNA-sequencing,
RT-qPCR,
Northern
blot
verification
experiments
on
to
explore
involvement

Summary
Nannochloropsis
oceanica
is
an
industrially
relevant
marine
microalga
rich
in
eicosapentaenoic
acid
(EPA,
a
valuable
ω‐3
polyunsaturated
fatty
acid),
yet
the
algal
production
potential
remains
to
be
unlocked.
Here
we
engineered
N.
synthesize
high‐value
carotenoid
astaxanthin
independent
of
high‐light
(HL)
induction
for
achieving
multifaceted
benefits.
By
screening
β‐carotenoid
ketolases
and
hydroxylases
various
origins,
strategically
manipulating
compartmentalization,
fusion
patterns,
linkers
enzyme
pair,
remarkable
133‐fold
increase
content
was
achieved
.
Iterative
metabolic
engineering
efforts
led
further
increases
synthesis
up
7.3
mg
g
−1
,
highest
reported
microalgae
under
nonstress
conditions.
Astaxanthin
found
photosystem
components
allowed
alga
HL
resistance
augmented
EPA
production.
Besides,
co‐production

fed‐batch
cultivation

untapped

light‐driven
chassis
