Haematococcus pluvialis is a precious algal species renowned for its ability to simultaneous production of astaxanthin and lipids. However, slow growth rate necessitates the development appropriate mutagenesis methodologies effectively enhance synchronous both This study introduced implemented co-mutation method Atmospheric Room Temperature Plasma(ARTP) ethanol. The process, performance, preliminary mechanisms underlying combined accumulation lipids in under influence ethanol combination with ARTP-induced mutation were compared analyzed. Findings showed that lipid contents relative total cell mass 110-2 strain reached 54.37%, surpassing 0-3 control by 16.98% 47.58% respectively. transcriptome-level analysis provided further insights into how ARTP induction could promote expressions carotenoid synthesis genes, thereby augmenting related enzymatic activities.

In eukaryotes, microRNAs (miRNAs) are generated by Dicer/Dicer-Like (DCL)-mediated cleavage. Previous studies identified three DCL genes (CrDCL1-3) in Chlamydomonas reinhardtii and indicated that CrDCL3 mediated the production of most miRNAs, while CrDCL1 protein was mainly involved siRNA biogenesis. The role miRNA biogenesis remains unclear. This study constructed a phylogenetic tree, performed structural analyses Dicer/DCL proteins from multiple species screened verified dcl1 dcl3 mutant strains. Using CC-5325 as control groups, we sRNA-sequencing, RT-qPCR, Northern blot verification experiments on to explore involvement C. reinhardtii. results demonstrated 22 including cre-miR910, novel-miR01, novel-miR03, novel-miR04, novel-miR05, novel-miR06, whose depended not only but also CrDCL3. present findings highlight some miRNAs may be CrDCL proteins, which is different animals plants. this will enrich knowledge eukaryotes.

Summary Nannochloropsis oceanica is an industrially relevant marine microalga rich in eicosapentaenoic acid (EPA, a valuable ω‐3 polyunsaturated fatty acid), yet the algal production potential remains to be unlocked. Here we engineered N. synthesize high‐value carotenoid astaxanthin independent of high‐light (HL) induction for achieving multifaceted benefits. By screening β‐carotenoid ketolases and hydroxylases various origins, strategically manipulating compartmentalization, fusion patterns, linkers enzyme pair, remarkable 133‐fold increase content was achieved . Iterative metabolic engineering efforts led further increases synthesis up 7.3 mg g −1 , highest reported microalgae under nonstress conditions. Astaxanthin found photosystem components allowed alga HL resistance augmented EPA production. Besides, co‐production by through fed‐batch cultivation approach. Our findings unveil untapped as robust, light‐driven chassis constitutive provide feasible strategies concurrent multiple biochemicals from CO 2 thereby paving way sustainable biotechnological applications this alga.

To explore the relationship between unsaturated fatty acid (UFA) content and parameters for microwave extraction, multimaterial multiparameter testing was conducted in which five kinds of materials with different UFA contents (potato, wheat, corn, soybean, peanut) were selected experiment. Then, optimization results synthesized Origin 8.0 software, fitting degree cubic model highest, determination coefficients 0.9984, 0.9991, 0.8953, 0.9989, residual sums squares 0.2888, 0.1587, 0.8265, 0.1864, correlation are ideal. stability accuracy verified rice, coefficient predicted value actual 0.9998.
