Ferroptosis, an iron and reactive oxygen species (ROS)-dependent non-apoptotic type of regulated cell death, is characterized by a massive overload peroxidation polyunsaturated fatty acids (PUFAs), which finally results in death. Recent studies suggest that ferroptosis can influence carcinogenesis negatively therefore may be used as novel anti-cancer strategy. Hepatocellular carcinoma (HCC) deadly malignancy with poor chances survival the second leading cause cancer deaths worldwide. Diagnosis at already late stage general resistance to current therapies responsible for dismal outcome. As liver acts key factor metabolism, shown play important role HCC and, more importantly, hold potential eradicate HCC. In this review, we summarize knowledge have application therapy option provide overview translation clinical practice

Abstract Background Stroke is a devastating disease affecting populations worldwide and the primary cause of long-term disability. The inflammatory storm plays crucial role in progression stroke. In acute phase ischemic stroke, there transient increase anti-inflammatory M2 microglia followed by rapid decline. Due to abundant phospholipid brain tissue, lipid peroxidation notable characteristic ischemia/reperfusion (I/R), constituting structural foundation for ferroptosis microglia. Slowing down decrease numbers controlling microenvironment holds significant potential enhancing stroke recovery. Results We found that inhibitor can modulate response MCAO mice, characterizing level microglia-related cytokines was increased. then confirmed different subtypes exhibit distinct sensitivities I/R-induced ferroptosis. Adipose-derived stem cells derived exosome (ADSC-Exo) effectively decreased susceptibility via Fxr2/Atf3/Slc7a11, suppressing promoting neuronal survival. Furthermore, through plasmid engineering, more efficient microglia-targeted exosome, termed M2pep-ADSC-Exo, developed. vivo vitro experiments demonstrated M2pep-ADSC-Exo exhibits targeting specificity microglia, further inhibiting improving neurological function mice. Conclusion Collectively, we illustrated novel therapeutic mechanism Fxr2 ADSC-Exo could alleviate regulating Atf3/Slc7all expression, hence microenvironment, neurofunction recovery cerebral I/R injury. obtained engineered modification, which improved capabilities toward This provides new avenue treatment Graphical

Ferroptosis has important value in cancer treatment. It is significant to explore the new ferroptosis-related lncRNAs prediction model Hepatocellular carcinoma (HCC) and potential molecular mechanism of lncRNAs. We constructed a prognostic multi-lncRNA signature based on differentially expressed HCC. qRT-PCR was applied determine expression lncRNA HCC cells. The biological roles NRAV vitro vivo were determined by performing series functional experiments. Furthermore, dual-luciferase reporter RNA immunoprecipitation (RIP) assays used confirm interaction with miR-375-3P. identified 6 differently associated prognosis Kaplan-Meier analyses revealed high-risk poor Moreover, AUC showed utility predicting prognosis. Further experiments show that high can strengthen viciousness Interestingly, we found enhance iron export ferroptosis resistance. study competitively binds miR-375-3P attenuates inhibitory effect SLC7A11, affecting patients In conclusion, developed novel predictive for induction through miR-375-3P/SLC7A11 axis.

Hepatocellular carcinoma is the most common form of primary liver cancer and poses a significant challenge to medical community because its high mortality rate. In recent years, ferroptosis, unique cell death, has garnered widespread attention. Ferroptosis, which characterized by iron-dependent lipid peroxidation mitochondrial alterations, closely associated with pathological processes various diseases, including hepatocellular carcinoma. Long non-coding RNAs (lncRNAs), are type functional RNA, play crucial regulatory roles in variety biological processes. this manuscript, we review lncRNAs key aspects summarize research progress on ferroptosis-related

Recently, immunotherapy combined with targeted therapy has significantly prolonged the survival time and improved quality of life patients hepatocellular carcinoma (HCC). However, HCC treatment remains challenging due to high heterogeneity this malignancy. Sorafenib, first-line drug for HCC, can inhibit progression by inducing ferroptosis. Ferroptosis is associated formation an immunosuppressive microenvironment in tumours. Moreover, long non-coding RNAs (lncRNAs) are strongly ferroptosis HCC. Discovery ferroptosis-related lncRNAs (FR-lncRNAs) critical predicting prognosis effectiveness therapies improve duration patients. Herein, all cases from The Cancer Genome Atlas (TCGA) database were divided into training testing groups at a 6:4 ratio construct validate lncRNA signatures. Least Absolute Shrinkage Selection Operator (LASSO) regression Cox analyses used screen six FR-lncRNAs (including MKLN1-AS, LINC01224, LNCSRLR, LINC01063, PRRT3-AS1, POLH-AS1). Kaplan-Meier (K-M) receiver operating characteristic (ROC) curve demonstrated optimal predictive prognostic ability signature. Furthermore, nomogram indicated favourable discrimination consistency. For further validation, we real-time quantitative polymerase chain reaction (qRT-PCR) analyse expression POLH-AS1 tissues. determined signature predict effects Gene set enrichment analysis (GSEA) somatic mutation showed that pathways, immune-related TP53 mutations may be overall (OS) outcomes Overall, our study suggests new risk model significant value it could contribute precise individualised treatment.

Liver cancer is one of the most common malignancies, with severe morbidity and mortality. While considerable progress has been made in liver treatment, 5-year overall survival (OS) patients not improved significantly. Reasons include inadequate capability early screening diagnosis, a high incidence recurrence metastasis, degree tumor heterogeneity, an immunosuppressive microenvironment. Therefore, identification validation specific robust biomarkers are major importance for screening, timely accurate prognosis, prevention progression. In this review, we highlight some latest research potential applications biomarkers, describing hotspots prospective directions biomarker discovery.
