Colorectal cancer (CRC) is a serious threat to human health with the third morbidity and second cancer-related mortality worldwide. It urgent explore more effective strategy for CRC because of acquired treatment resistance from non-surgical conventional therapies, including radiation, chemotherapy, targeted therapy immunotherapy. Ferroptosis novel form programmed cell death characterized by iron-dependent lipid peroxidation species (ROS) accumulation has been identified as promising target treatment, especially those resistance. In this review, we mainly summarize recent studies on influence regulation ferroptosis which (including gut microbiota) modulating metabolism iron, amino acid lipid. Thus analysis may provide potential targets inducing shed lights future application in CRC.

Ferroptosis is a recently discovered pattern of programmed cell death that nonapoptotic and irondependent. It involved in lipid peroxidation dependent on reactive oxygen species. has been verified to play crucial regulatory role variety pathological courses disease, particularly cancer. Emerging research highlighted the potential ferroptosis tumorigenesis, cancer development resistance chemotherapy. However, mechanism remains unclear, which limits application treatment. Noncoding RNAs (ncRNAs) are noncoding transcripts regulate gene expression various ways affect malignant phenotypes cells. At present, biological function underlying ncRNAs have partially elucidated. Herein, we summarize current knowledge central network ferroptosis, with focus functions ferroptosis. The clinical prospects ferroptosis-related diagnosis, prognosis anticancer therapies also discussed. Elucidating along assessing significance ncRNAs, provides new perspectives for understanding biology treatment approaches, may benefit numerous patients future.

LncRNA is a type of transcript with length exceeding 200 nucleotides, which was once considered junk no biological function during the transcription process. In recent years, lncRNA has been shown to act as an important regulatory factor at multiple levels gene expression, affecting various programmed cell death modes including ferroptosis. Ferroptosis, new form death, characterized by deficiency cysteine or inactivation glutathione peroxidase, leading depletion glutathione, aggregation iron ions, and lipid peroxidation. These processes are influenced many physiological processes, such Nrf2 pathway, autophagy, p53 pathway so on. An increasing number studies have that can block expression specific molecules through decoy effect, guide proteins function, promote interactions between scaffolds. action regulate key factors in metabolism, antioxidant metabolism epigenetic genetic regulation, thereby regulating process this review, we snapshotted mechanism ferroptosis example, emphasizing regulation on these pathways, helping fully understand evolution fate.

Background Long noncoding RNAs (lncRNAs) have emerged as critical regulators of the expression genes involved in cardiovascular diseases. This project aims to identify circulating lncRNAs associated with protein‐coding mRNAs differentially expressed between hypertensive and normotensive individuals establish their link hypertension. Methods Results The analyses were conducted 3 main steps: (1) an unbiased whole blood transcriptome‐wide analysis was replicate by hypertension status 497 179 Black from GENE‐FORECAST (Genomics, Environmental Factors Social Determinants Cardiovascular Disease African‐Americans Study) MH‐GRID (Minority Health Genomics Translational Research Bio‐Repository Database) studies, respectively. Subsequently, (2) proximal lncRNAs, termed cis lncRNA quantitative trait loci, each mRNA identified study replicated study. Finally, (3) loci used predictors a random forest model predict both data sets. A total 129 significantly lncRNA‐mRNA association revealed 249 102 mRNAs, including VAMP2 (vesicle‐associated membrane protein 2), mitogen‐activated kinase 3, CCAAT enhancer binding beta, lymphocyte antigen 6 complex, locus E. predicted area under curve 0.79 0.71 Conclusions leveraged significant sample individuals, population facing disproportionate burden unveiled 271 relationships involving that play roles vascular pathways relevant pressure regulation. compelling findings, consistent across 2 independent sets, reliable foundation for designing vitro/in vivo experiments.

This study analysed relationship between protein ameloblastin (AMBN) potential biological markers associated with TGCT diagnosis prognosis. The AMBN was determined by bioinformatic analysis using expression profiles three RNAs (long non-coding (lncRNAs), microRNAs (miRNAs) mRNAs) from Cancer Genome Atlas (TCGA) database, available clinical information corresponding patients. Prediction validation competitive endogenous RNA (ceRNA) regulatory networks related to performed. its ceRNA network were found be recurrence TGCT, LINC02701 may used as diagnostic factor TGCT. Furthermore, we identified PELATON (Plaque Enriched LncRNA In Atherosclerotic And Inflammatory Bowel Macrophage Regulation) an independent prognostic for progression-free interval.

Emerging insights into iron-dependent form of regulated cell death ferroptosis in cancer have opened a perspective for its use therapy. Of interest, systematic profiling gene signatures as prognostic factors has gained special attention several cancers. Herein, we sought to investigate the presence repetitive genomes vicinity genes that may influence their expression and establish signature associated with multiple myeloma (MM). Our analysis showed were enriched genome vicinity, strong predominance SINE family, followed by LINE, which most significant discriminant values SINE/Alu LINE/L1, respectively. In addition, examined detail performance these risk prediction model specified fourteen ferroptosis-related signatures, identified MM high-risk patients lower immune/stromal scores higher tumor purity immune microenvironment. also found lncRNA CRNDE correlated score was highly majority comprising signature. Taken together, propose molecular impact highlighted on local transcriptome cancer. Furthermore, revealed genomic signature/biomarker related can be used predict survival patients.
