We integrated single-cell RNA sequencing (scRNA-seq) transposase-accessible chromatin (scATAC-seq) data from multi-omics perspective. Based on consensus non-negative matrix factorization (cNMF) algorithm, functionally heterogeneous cancer cells were classified into metabolism, inflammatory, EMT meta programs, with spatial transcriptomics (stRNA-seq) providing information such disparate programs cells. bulk (RNA-seq) revealed high clinical prognostic values three transcription factor regulatory network motif activities revealing key factors that regulate interactions between varying other subpopulations microenvironment investigated. Patients with combined hepatocellular-cholangiocarcinoma (cHCC-CCA) have limited treatment options and poor prognosis. Tumor-associated macrophages (TAMs) are the most abundant infiltrating immune cells in tumor microenvironment promote stemness, proliferation, invasion metastasis. Evidence suggested that transthyretin (TTR) influenced prolifetation functions of different tumors play an essential role microenvironment.

Cholangiocarcinoma (CCA) is an extremely aggressive malignancy characterized by a very limited prognosis and scarce treatment options. The majority of patients are diagnosed at advanced stage do not qualify for potentially curative surgical treatments, making CCA increasingly prevalent global challenge. highly reactive desmoplastic stroma, with complex mechanisms underlying the mutual interactions between tumor cells stromal compartment. This review focuses on recent studies examining CCA’s biological features, particular reference to stroma (TRS) its role in progression, including matrix remodeling, angiogenesis lymphangiogenesis, metastasis, immune evasion. After giving panoramic view relationship tumoral compartment (cancer-associated fibroblast, CAFs tumor-associated macrophages, TAMs), this also discusses current therapeutic approaches counteract TAMs effects progression.

OBJECTIVE: This article aims to clarify pitfalls and find strategies for the detecting diagnosing hyperechoic liver metastases (LMs) using Sonazoid-contrast enhanced ultrasonography (Sonazoid-CEUS). METHODS: study was a prospective self-controlled study. Patients with hepatic lesions suspected as LMs or benign were included in Baseline (BUS) Sonazoid-CEUS performed on every patient. Characteristics of nodules compared by chi-square test fisher test. Factors influenced CEUS demonstrated univariate analysis multivariate logistic regression analysis. RESULTS: 54 patients this found additional 75 from 19 Kupffer phase. We focal deep seated are main confounding factors diagnosis. Sensitivity would be improved 16.67% 78.57%, negative predictive value (NPV) 28.57% 76.92% accuracy 37.5% 87.50% when rapid “wash-in” “wash-out” diagnostic criteria. CONCLUSIONS: Hyperechoic especially deeply ones usually not shown typical “black hole” sign Quickly “wash-in wash out” shows high malignant nodules. highly recommend routing exam detect diagnose LMs.

Spatial transcriptomics is revolutionising the exploration of intratissue heterogeneity in cancer, yet capturing cellular niches and their spatial relationships remains challenging. We introduce SpottedPy, a Python package designed to identify tumour hotspots map interactions within cancer ecosystem. Using we examine epithelial-mesenchymal plasticity breast highlight stable associated with angiogenic hypoxic regions, shielded by CAFs macrophages. Hybrid mesenchymal hotspot distribution followed transformation gradients reflecting progressive immunosuppression. Our method offers flexibility explore at different scales, from immediate neighbours broader tissue modules, providing new insights into microenvironment dynamics.
