Abstract Sepsis is the leading systemic inflammatory response syndrome in worldwide, yet relatively little known about genes and signaling pathways involved sepsis progression. The current investigation aimed to elucidate potential key candidate its associated complications. Next generation sequencing (NGS) dataset (GSE185263) was downloaded from Gene Expression Omnibus (GEO) database, which included data 348 samples 44 normal control samples. Differentially expressed (DEGs) were identified using t-tests DESeq2 R package. Next, we made use of g:Profiler analyze gene ontology (GO) REACTOME pathway. Then protein-protein interaction (PPI) these DEGs visualized by Cytoscape with Search Tool for Retrieval Interacting Genes (STRING). Abstract Objectives Colorectal cancer (CRC) is a global challenge, and heat shock protein 90 (HSP90) identified as key driver in progression. However, the tumor-promoting mechanism of HSP90 CRC, particularly HSP90AB1, remains unclear. This study aims to explore analyze oncogenic HSP90AB1 CRC identify potential therapeutic targets. Materials methods expression underwent analysis cell lines tissues at mRNA levels. Through use shRNA, targeted suppression was achieved lines, enabling its influence on proliferation, invasion, apoptosis, cycle Subsequent investigation focused elucidating regulatory relationship between IDO1, employing combination bioinformatics approaches vitro /vivo experiments. These efforts confirmed IDO1 downstream target provided insight into role driving Results exhibits overexpression both tumor (p<0.05). Its downregulation impedes proliferation invasion (p<0.01), promotes apoptosis arrest Investigation reveals that decreased leads inhibition suggesting regulation plays crucial mediating pro-tumorigenic effects HSP90AB1. In vivo experiments confirm substantial reduction growth upon knockdown xenograft models (p<0.01). this tumor-suppressive effect reversed highlighting Conclusions exerts progression by upregulating IDO1.
