The Wnt signaling pathway plays a crucial role in development and tissue homeostasis, regulating key cellular processes such as proliferation, differentiation, apoptosis. However, its abnormal activation is strongly associated with tumorigenesis, metastasis, resistance to therapy, making it vital target for cancer treatment. This review provides comprehensive insight into the of cancer, examining normal physiological functions, dysregulation malignancies, therapeutic potential. We emphasize importance predicting sensitivity identify biomarkers across various types. Additionally, we address challenges future prospects Wnt-targeted therapies, including biomarker discovery, advancements emerging technologies, their application clinical practice.

Cancer biomarker discovery is essential for early detection and monitoring, yet there a lack of comprehensive studies examining organ-specific biomarkers across various cancer types. In this study, we analyzed clinical data from 59,184 patients diagnosed between 2013 2023, focusing on 11 major systems. We used propensity score matching with 55,010 healthy controls to create balanced comparison groups. Serum profiles were assessed through principal component analysis, differential expression ROC curve analysis. Our findings revealed patterns, such as decreased CA724, ferritin, β2-microglobulin in thoracic cancer, reduced serum phosphorus neurological elevated cystatin C creatinine urinary system cancer. Further analysis 22 types uncovered additional biomarkers, including ALT hepatobiliary altered coagulation factors laryngeal increased monocytes pancreatic complement C3 intestinal These results provide valuable insights into the unique signatures different cancers, contributing potential development more targeted efficient screening methods.

Ovarian cancer (OC) is the most common cause of death in female cancers. The prognosis OC very poor due to delayed diagnosis and identification patients advanced stages, metastasis, recurrence, resistance chemotherapy. As chemotherapy with platinum-based drugs such as cisplatin (DDP) main treatment cases, DDP an important obstacle achieving satisfactory therapeutic efficacy. Consequently, knowing different molecular mechanisms involved necessary achieve new approaches. According numerous recent studies, non-coding RNAs (ncRNAs) could regulate proliferation, differentiation, apoptosis, chemoresistance many cancers, including OC. Most these ncRNAs are released by tumor cells into human fluid, allowing them be used tools for diagnosis. Dysregulated expression of specific non-coding RNAs (ncRNAs) has been strongly linked to tumorigenesis, cancer progression, and therapeutic resistance. These ncRNAs can act as either oncogenes or tumor suppressors, thereby serving valuable diagnostic prognostic markers. Numerous studies have implicated the participation in regulation diverse signaling pathways, including pivotal Wnt/β-catenin pathway that is widely acknowledged for its role embryogenesis, cellular proliferation, biology control. Recent emerging evidence shed light on capacity interact with key components pathway, modulating Wnt target genes cells. Notably, activity this reciprocally influence levels ncRNAs. However, comprehensive analysis investigating associated their intricate interactions remains elusive. Based these noteworthy findings, review aims unravel associations between during initiation, potential implications interventions. Additionally, we provide a overview characteristics accompanied by thorough discussion functional roles biology. Targeting molecules may emerge promising effective strategy future treatments.

Colorectal cancer (CRC) development involves a series of molecular events that drive the progression from normal colorectal epithelium to adenoma and eventually invasive carcinoma. While involvement extrachromosomal circular DNAs (ecDNAs) in genome remodeling has been established, their specific roles CRC formation remain unclear. Using Circle-Sequencing whole transcriptomic sequencing, we comprehensively profile transcriptomes healthy individuals, adenoma, patients. Our delineate analyses characterize key involved oncogene expression through normal-adenoma–carcinoma continuum highlight immune response-related pathways cell cycle pathways, are dominat progression.
