Ulcerative colitis (UC) is a chronic inflammation disease with severe impact on quality of life, limited treatment options. Ramulus Mori alkaloids (SZ-A) from Morus alba show promise for UC due to their safety and pharmacological effects, including anti-inflammation barrier repair. However, clinical use has been by gastrointestinal flatulence as side effect action an α-glucosidase inhibitor targeting the small intestine following oral administration. Therefore, constructing colon-targeted formulation deliver SZ-A advantageous strategy improve therapy. In this study, we used complex formed thiolated hyaluronic acid, which mucosal adhesion inflammation-targeting properties, intermediate carrier prepared sodium alginate-modified PLGA microspheres (SZ-A@MSs) double emulsion method achieve efficient encapsulation SZ-A. Specifically, alginate serves gastric acid protectant microbiota-responsive material, enabling precise responsive release in colonic region. SZ-A@MSs have particle size about 30 µm, drug loading 12.0%, efficiency 31.7% function through intestinal inflammatory sites. showed antioxidant anti-inflammatory abilities Raw264.7 cells. vivo imaging results suggest that good colon site retention sustained-release effect.
