In recent years, the incidence of neurodegenerative diseases, such as Alzheimer’s disease, Parkinson’s Huntington’s and amyotrophic lateral sclerosis, has shown a steadily rising trend, which posed major challenge to global public health. Traditional Chinese Medicine (TCM), with its multi-component multi-target characteristics, offers promising approach for treatment diseases. However, it is difficult comprehensively elucidate complex mechanisms underlying TCM formulations. Exposure to patulin (PAT) poses a significant health risk animals, emphasizing the need for natural, safe substances mitigate toxicity. Acremonium terricola culture (ATC), fungal fermentation-derived feed additive, is known its antioxidant properties, yet potential alleviate mycotoxin-induced toxicity remains largely uninvestigated. In this study, ethanol extracts from ATC (EEAT) were prepared with total phenolic content of 67.9 mg GAE/g and flavonoid 32.7 RE/g. Ultra-high performance liquid chromatography coupled tandem mass spectrometry (UPLC-ESI-MS/MS) analysis was employed investigate bioactive components in EEAT. PAT-exposed Caenorhabditis elegans models, EEAT treatment significantly enhanced locomotory capacity elevated enzyme activities by 63.1% (SOD) 90.1% (GSH-ST), respectively. Molecular docking revealed that key active compounds EEAT, such as coumarin, succinic acid, trigonelline, exhibited effective binding affinities targets SIR-2.1 DAF-2. Notably, coumarin trigonelline most alleviating PAT toxicity, evidenced rescued locomotor rates oxidative impairment C. elegans. Our findings not only elucidate molecular basis EEAT-mediated mitigation but also establish A. sustainable antioxidant.
