Bletilla striata, a medicinal orchidaceous plant, is recognized for its significant pharmacological value. However, the lack of comparative metabolomic data across different extraction methods analyzing bioactive components has significantly undervalued application potential B. striata in traditional Chinese medicine market. Using six ultrasound-assisted and UPLC-MS/MS, this study identified 1,945 metabolites extracts. The dominant categories were lipids (51.35%), flavonoids (18.00%), phenolic acids (12.51%). KEGG analysis revealed alterations isoflavonoids biosynthesis pathways. Thirteen bitter metabolites, including cinnamic acid, tubers, underscoring their applications, such as anti-inflammatory, antioxidant antibacterial activities. Optimizing can better preserve extracts, thereby enhancing applications food pharmaceutical industries.

Microglia, the resident immune cells of central nervous system, continuously monitor brain’s microenvironment through their array specific receptors. Once brain function is altered, microglia are recruited to sites perform functions, including phagocytosis misfolded proteins, cellular debris, and apoptotic maintain homeostasis. When toxic substances overproduced, over-activated produce large amounts pro-inflammatory cytokines, which induce chronic inflammatory responses lead neurotoxicity. Additionally, can also protect neuronal microglia-neuron crosstalk. Microglia receptors important mediators for receive external stimuli, regulate functional state microglia, transmit signals between cells. In this paper, we first review role microglia-expressed in pathogenesis treatment Alzheimer’s disease; moreover, emphasize complexity targeting therapeutic interventions neurodegenerative disorders inform discovery new biomarkers development innovative therapeutics

ABSTRACT Diabetic nephropathy (DN) is the most common cause of end‐stage renal disease. Its pathogenesis includes inflammation, an excess reactive oxygen species, and kidney damage. The present study intended to explore nephroprotective effects acacetin (ACN) in streptozotocin‐induced diabetic animals. following are experimental groups: One millilitre 0.9% saline was given Group I (control), Streptozotocin (STZ) (diabetic animals) + II (DN group) (negative control), DN ACN (15 mg/kg body weight [bw]) III, Valsartan (150 bw) IV. According findings, decreased levels glucose, serum creatinine (Scr), blood urea nitrogen (BUN), malondialdehyde (MDA), proinflammatory cytokines while increasing bw, superoxide dismutase (SOD), glutathione peroxidase (GSH‐Px) comparison histopathological analysis revealed that animals treated with showed recovery damage tissues caused by STZ. In STZ‐induced mice, reduced upregulating proteins 5′ adenosine monophosphate‐activated protein kinase (AMPK), p‐AMPK, SIRT1 downregulating TGF‐β, COL‐1, COL‐IV, NF‐κB, ASC, NLRP3, GSDMD, according western blot analysis. Hence, current demonstrated regulation AMPK/SIRT1 NF‐κB/ASC/NLRP3 inflammasome pathways mice responsible for protective ACN. may therefore be a viable treatment option DN.

Globally, cardiovascular disease (CVD) has emerged as a leading cause of mortality and morbidity. As the world’s population ages, CVD incidence is on rise, extensive attention been drawn to optimizing therapeutic regimens. Acacetin, natural flavonoid derived from various plants, demonstrated have wide spectrum pharmacological properties, such antioxidant, anti-inflammatory, anti-bacterial, anti-tumor activities, well protective effects diverse tissues organs. Recently, increasing numbers studies (mostly preclinical) indicated that acacetin potential might become novel strategy for CVDs. The importance in treatment necessitates systematic comprehensive review its system underlying mechanisms involved. Here, we first provide an overview some basic properties acacetin. Subsequently, multiple CVDs, like arrhythmias, cardiac ischemia/reperfusion injury, atherosclerosis, myocardial hypertrophy fibrosis, drug-induced cardiotoxicity, diabetic cardiomyopathy, hypertension, senescence, are discussed detail. by which exhibits protection appear involve suppressing oxidative stress, reducing inflammation, preventing cardiomyocyte apoptosis endothelial cell regulating mitochondrial autophagy lipid metabolism. Meanwhile, several critical signaling pathways also found mediate against including phosphoinositide 3-kinase/protein kinase B/mechanistic target rapamycin (PI3K/Akt/mTOR), sirtuin 1/AMP-activated protein kinase/peroxisome proliferator-activated receptor-γ coactivator-1α (Sirt1/AMPK/PGC-1α), transforming growth factor-β1/small mothers decapentaplegic 3 (TGF-β1/Smad3), B/endothelial nitric oxide synthase (Akt/eNOS), others. Finally, highlight existing problems associated with need be addressed, requirement clinical evidence enhanced bioavailability, promising drug candidate.

The pyroptosis of cardiomyocytes has become an essential topic in heart failure research. abnormal accumulation these biological factors, including angiotensin II, advanced glycation end products, and various growth factors (such as connective tissue factor, vascular endothelial transforming factor beta, among others), activates the nuclear factor-κB (NF-κB) signaling pathway cardiovascular diseases, ultimately leading to cardiomyocytes. Therefore, exploring underlying molecular mechanisms is for developing novel drugs therapeutic strategies. However, our current understanding precise regulatory mechanism this complex cardiomyocyte still limited. Given this, study reviews milestone discoveries field research since 1986, analyzes detail similarities, differences, interactions between other cell death modes apoptosis, necroptosis, autophagy, ferroptosis), explores deep connection failure. At same time, it depicts complete activation, transmission, eventual NF-κB process In addition, also systematically summarizes approaches that can inhibit reduce pyroptosis, drugs, natural compounds, small molecule inhibitors, gene editing, cutting-edge technologies, aiming provide solid scientific support new perspectives prevention treatment
