Osteoarthritis (OA), caused by the long-term use of joints, is a representative degenerative disease in elderly. However, recently, age onset has been decreasing owing to excessive activities among young people their 20s and 30s. Gynostemma pentaphyllum (Thunb.) Makino (GP), perennial herb Cucurbitaceae family, used since Ming dynasty as medicinal material treat various ailments, such rheumatism, liver disease, diabetes. In this study, we investigated anti-arthritic effects heat-processed extract (Actiponin (AP)) its derivatives, damulin A (DA) B (DB), using vitro (primary rat chondrocytes SW1353 cells) vivo (destabilization medial meniscus (DMM)-induced OA model) systems. Histological analysis results from study showed that group underwent DMM surgery induced degeneration loss proteoglycan destruction cartilage (OARSI score 14 ± 0.57), whereas received AP daily for 8 weeks maintained an intact condition 5 0.28 at 200 mg/kg, p < 0.001). addition, thickness were reduced group, but restored AP-administered group. Furthermore, von Frey pain threshold was considerably low (54.5 g weeks), dose-dependently increased (65.5, 69.5, 70.3, 71.8 30, 50, 100, respectively). studies AP, DA, DB expression interleukin-1β alone-induced nitrite; inducible nitric oxide synthase; cyclooxygenase-2; matrix metallopeptidase 1/3/13; disintegrin metalloproteinase with thrombospondin motifs 4/5. They also collagen type II aggrecan, which are components extracellular matrix. The confirmed be mediated mitogen-activated protein kinase nuclear factor kappa-light-chain-enhancer activated cell signaling pathways. Collectively, these suggest potential therapeutic agent mitigating progression chondroprotection.
