Andrographis paniculata is a plant of the Acanthaceae family and its primary bioactive constituent, andrographolide, exhibits broad spectrum pharmacological activities notable clinical efficacy. However, poor solubility limited bioavailability pose significant challenges for therapeutic applications. To overcome these limitations, researchers have synthesized andrographolide sulfonates by reacting with ethanol sulfuric acid. This sulfonated derivative significantly enhances water while retaining key properties such as anti-inflammatory antiviral activities. As representative formulation, Xiyanping injection has been widely employed in treatment respiratory infections, pneumonia, related conditions, playing critical role during COVID-19 pandemic. Despite widespread application, there yet to be comprehensive review chemical composition mechanisms. Additionally, safety remains topic some debate. systematically examines composition, activities, applications, adverse reactions their formulation provide scientific basis further research also offering valuable insights development similar drugs.

Andrographolide (AP) has been shown to possess anti-inflammatory activities. In this study, the impact of AP in sepsis-induced acute liver injury (ALI) and molecules involved were dissected. FKBP1A was predicted be sole target protein that also differentially expressed GSE166868 dataset. induced expression suppressed NOTCH1 a dose-dependent manner. ameliorated ALI mice by D-galactosamine LPS inhibited LPS-induced parenchymal cell vitro. By contrast, protective effect significantly lost after knockdown FKBP1A. As positive control, therapeutic dexamethasone on may related NOTCH1, which not promoted AK2 transcription cells, endoplasmic reticulum (ER) stress impairing NOTCH1/AK2 signaling. Restoration reversed hepatoprotective activating ER pathway. Therefore, AP-promoted inhibits progression blocking NOTCH1/AK2-mediated
