Autism is a neurodevelopmental disorder with limited treatment alternatives and which incidence increasing. Some research suggests that vagus nerve simulation might lead to the reduction of certain symptom. Therefore, we aimed examine effect bilateral transcutaneous auricular stimulation (tVNS) on inflammatory response in an adult valproic acid (VPA) induced mouse (C57BL6) model autism for first time. The was by oral VPA administration (600 mg·kg −1 ) C57BL/6 pregnant mice E12.5 days. study included three groups: Transcutaneous Auricular Stimulation Group (VPA + tVNS), Control sham), Healthy (Control sham). Each group 16 (8 M/8 F). Our results show serum IL‐1β IL‐6 levels were significantly higher male VPA‐exposed than controls. However, lower, IL‐6, TNF‐ α, IL‐22 not different female compared control group. Brain NLRP3 both sexes ( P < 0.05). tVNS application increased brain reduced mice. We conclude cytokine dysregulation associated VPA‐induced model, more pronounced altered NLPR3 sexes. Further studies are needed understand beneficial or detrimental role its sexual dimorphism.
