Romboutsia is a dominant genus in the goose intestine. Recent studies have suggested that lituseburensis might regulate serum immunoglobulin levels female geese, although underlying mechanisms remain unclear. In this study, we administered (R. lituseburensis) orally to leading successful colonization of ileum. Subsequent analysis showed IgM, IgA, and IgG significantly decreased after (P < 0.01). 16S rRNA sequencing revealed R. altered microbial composition increased relative abundance Jeotgalicoccus 0.01), Turicibacter, Bacillus 0.05) Transcriptome further identified 263 differentially expressed genes (DEGs) ileum (146 upregulated, 117 downregulated) 725 DEGs spleen (300 425 downregulated). Kyoto Encyclopedia Genes Genomes (KEGG) pathway enrichment these were enriched 17 pathways 21 spleen. Notably, "Intestinal immune network for IgA production" was 0.05). Further, Short Time-series Expression Miner (STEM) grouped 2 tissues into 49 clusters, with clusters 27 29 showing highest significance similar expression patterns. Pathway confirmed both clusters. Furthermore, protein-protein interaction (PPI) along correlation between microbiota gene expression, highlighted KEL, SERPING1, CALR, OSTN as key hub genes. Overall, Jeotgalicoccus, Concurrently, it downregulate (CCR9, TNFRSF13B, AICDA) via OSTN, thereby contributing reduction levels. These findings offer new insights how influences function geese provide theoretical basis research its other physiological roles geese.
