Dirigent (DIR) proteins are key regulators of lignin and lignan biosynthesis play critical roles in plant hormone responses, abiotic stress tolerance, growth development. This study identified characterized 47 PeDIR genes Moso bamboo, classifying them into three groups. Phylogenetic comparative analyses revealed strong evolutionary conservation, with the bamboo being most closely related to those rice maize. DIR within each subfamily exhibited high conservation motif composition, domain structure, 3D configuration. Subcellular localization protein interaction studies further elucidated gene functions. Specifically, PeDIR02 primarily localized cell membrane was shown be unable form homodimers yeast two-hybrid (Y2H) assays. Transcriptome expression involvement rapid shoot growth, indicating wall modification. qRT-PCR data also demonstrated responsiveness these hormones stresses, such as drought salinity. constructed first comprehensive regulatory network between transcription factors (TFs) genes, identifying ERF, DOF, MYB TFs synergistic expression.
