Identifying gene that enhances specific product accumulation of industrial strains is a central tenet metabolic engineering. Aurantiochytrium sp. has come into the limelight by virtue their biotechnological importance in production bioactive compounds, including fatty acids and carotenoids. However, there restrict high yield carotenoids This study, KAS Ⅲ-like enzyme 'YxwZ3′ were successfully used to construct recombinant cassette. We utilized TEF1 EM7 promoters overexpress YxwZ3 SZU445. The transformed strain displayed 4.88-fold increase total compared wild-type strain, involved β-carotene (37.96 μg/g), phytoene (1.55 β-cryptoxanthin (1.57 antheraxanthin (0.02 α-cryptoxanthin (0.25 μg/g) echinenone (0.72 with downregulated. It speculated more malonyl-CoA was MVA pathway for synthesis based on overexpression instead acid pathway. In addition, content lines increased 1.33 times (56.52 through optimization fermentation condition response surface methodology. These findings demonstrated significant impact sp., provided theoretical basis constructing high-yielding carotenoid-producing strains.

Staple grains with low levels of provitamin A carotenoids contribute to the global prevalence vitamin deficiency and therefore are main targets for biofortification. However, carotenoid stability during both seed maturation postharvest storage is a serious concern full benefits biofortified grains. In this study, we utilized Arabidopsis as model establish biofortification strategies in seeds. We discovered that manipulation biosynthetic activity by seed-specific expression Phytoene synthase (PSY) increases total but increased prone degradation storage, consistent previous studies contrast, stacking Orange (ORHis ), gene initiates chromoplast biogenesis, dramatically enhances content stability. Up 65- 10-fold β-carotene carotenoids, respectively, composing over 63% were observed seeds containing ORHis PSY. Co-expression Homogentisate geranylgeranyl transferase (HGGT) PSY further accumulation storage. Moreover, knocking-out hydroxylase 2 (BCH2) CRISPR/Cas9 not only potentially facilitates also minimizes negative effect production on germination. Our findings provide new insights into various processes multiplexed strategy simultaneously target biosynthesis, turnover, stable crop seeds. The online version contains supplementary material available at 10.1007/s42994-021-00046-1.
