Traumatic brain injury (TBI) has emerged as an increasing public health problem but not been well studied, particularly the mechanisms of cellular behaviors during TBI. In this study, we established ischemia/reperfusion (I/R) mice model using transient middle cerebral artery occlusion (tMCAO) strategy. After then, RNA-sequencing frontal lobes was performed to screen key inducers To further verify selected genes, collected peripheral blood mononuclear cells (PBMCs) from TBI patients within 24 h who attended intensive care unit (ICU) in Affiliated Hospital Yangzhou University and analyzed genes expression RT-qPCR. Finally, receiver operator characteristic (ROC) curves co-expression with senescence markers were applied evaluate predictive value genes. A total six screened out based on their novelty implications apoptosis signaling. RT-qPCR analysis PBMCs showed all up-regulated after comparing healthy volunteers hospital for physical examination. The area under ROC (AUC) >0.7, scores significantly positive. We thus identified TGM1, TGM2, ATF3, RCN3, ORAI1 ITPR3 novel that are induced TBI, these may also serve potential predictors progression
