Abstract Illuminated nights reduce melatonin peak and disrupt sleep. Using this as the basis of present experimental paradigm, we investigated whether nocturnal levels were crucial for sleep regulation in a diurnal vertebrate. Acclimated Indian house crows ( Corvus splendens ) randomly segregated into three groups 12 each. For next 10 days, one group was maintained on L:12 D, before (LD control); other two groups, absolute darkness replaced with dim light at night (dLAN; L = ~150 lux, D ~6 lux). Under dLAN, half an hour off time, LD control dLAN received intraperitoneally 200 μL vehicle (0.75% physiological saline), while similar but containing dose 50 μg bird −1 day . exogenous elevated AANAT mRNA plasma induced changes expressions clock genes PER2 , CRY1 BMAL1 NPAS2 REVERB pineal gland hypothalamus, encoding receptors MEL1a MEL1b epigenetic modifiers HAT1 HDAC2 HDAC4 DNMT3a hypothalamus. There have been concerns about the potential health risks posed by microplastics (MP). The detection of MP in a variety food products revealed that humans are ingesting MP. Nevertheless, there is paucity data their impacts, as well uptake, on intestinal barrier integrity. This study examined toxic effects oral administration two doses polyethylene (PE-MP) (3.75 or 15 mg/kg/day for 5 weeks; mean particle size: 4.0-6.0 µm) integrity rats. Moreover, effect melatonin treatment with exposure was also assessed. PE-MP histopathological changes, Alcian blue staining, Muc2 mRNA, proinflammatory cytokines (IL-1β and TNF-α), cleaved caspase-3, tight junction proteins (claudin-1, myosin light-chain kinase (MLCK), occludin, zonula occludens-1 (ZO-1)) were Oral resulted apparent jejunal alterations; significantly decreased mucin secretion, ZO-1, claudin-1 expression; upregulated MLCK IL-1β concentration, caspase-3 expression. Melatonin reversed these altered parameters improved PE-MP-induced ultrastructure changes. highlighted PE-MP's protective melatonin.

Melatonin (MLT) is a biologically active indoleamine involved in regulating various biological rhythms, which deficient individuals with Type 2 diabetes. The present study examined the effects of MLT on diabetic neuropathy (DN). Diabetic rats received treatment for 12 weeks, after changes kidney histology, oxidative damage, mitochondrial morphology and autophagy were measured. glucose tolerance‑ isoflurane tolerance‑area under curve (AUC) values relative renal weight index (RI) diabetes mellitus (DM) group significantly higher compared those control group. Abstract Worldwide trends to delay childbearing have increased parental ages at birth. Older age may harm offspring health, but mechanisms remain unclear. Alterations in DNA methylation (DNAm) patterns could play a role as aging has been associated with changes gametes of older individuals. We meta‐analyzed epigenome‐wide associations blood DNAm over 9500 newborns and 2000 children (5–10 years old) from the Pregnancy Childhood Epigenetics consortium. In newborns, we identified 33 CpG sites 13 loci maternal (P FDR < 0.05). Eight these CpGs were located near/in MTNR1B gene, coding for melatonin receptor. Regional analysis them together differentially methylated region consisting 9 in/near , which higher was greater = 6.92 × 10 −8 ) newborns. childhood samples, differences nominally significant ( p 0.05) retained same positive direction, suggesting persistence associations. The thyroid gland is the primary site of sodium/iodide symporter (NIS), an intrinsic plasma membrane protein responsible for active uptake iodine, which indispensable hormone synthesis. Since exposure to NIS inhibitors can potentially have harmful effects on entire organism, it important investigate potential protective known antioxidants, such as melatonin and indole-3-propionic acid (IPA), against pro-oxidative action classic inhibitors. study aimed check if what extent IPA interact with some confirmed regarding their oxidative damage lipids in thyroid. For comparison gland, typically present, liver tissue-not possessing NIS-was applied present study. Thyroid homogenates were incubated presence tested (i.e., NaClO3, NH4SCN, KSeCN, KNO3, NaF, KClO4, BPA) different ranges concentrations with/without (5 mM) or mM). malondialdehyde+4-hydroxyalkenals (MDA + 4-HDA) concentration (LPO index) was measured spectrophotometrically. NaClO3 increased LPO liver, but these not prevented by either IPA. Instead, NH4SCN observed both tissues indole substances. KSeCN NaF only thyroid, BPA did increase LPO, be due low resulting from restricted solubility. In conclusion, caused inhibitors, this indoleamine shoud considered a agent when produced appropriately living organisms also exogenous substance recommended individuals overexposed

The neurohormone melatonin (MLT) demonstrates promising potential in ameliorating neuropathic pain induced by paclitaxel (PTX) chemotherapy. However, little is known about its protective effect on dorsal root ganglion (DRG) neurons resulting from the chemotherapeutic drug PTX. Here, PTX-treated rats revealed that intrathecal administration of MLT dose-dependently elevated hind paw withdrawal thresholds and latency, indicating significantly reversed PTX-induced pain. Mechanistically, analgesic effects were found to be mediated via receptor 2 (MT2), as pretreatment with an MT2 antagonist inhibited these effects. Moreover, injection pNEK2-dependent epigenetic program All caused blocked receptor-selective antagonist, 4P-PDOT. Remarkably, multiple administered during PTX treatment (PTX+MLTs) exhibited not only rapid but also lasting reversal allodynia/hyperalgesia compared single-bolus after (PTX+MLT). In addition, PTX+MLTs greater efficacy reversing alterations pRSK2, pNEK2, JMJD3, H3K27me3, TRPV1 expression interaction DRG than PTX+MLT. These results indicated reduced incidence and/or severity neuropathy had a better inhibitory treatment. conclusion, MLT/MT2 therapy for painful chemotherapy may more effective prevention or reduction maintaining quality.
