Due to oxidative damage and mitochondrial dysfunction, boar semen cryopreservation remains a significant challenge. This study investigates the effects of pyrroloquinoline quinone (PQQ), mitochondrial-targeted antioxidant, on post-thaw sperm quality during cryopreservation. Boar was diluted in freezing extender containing different concentrations PQQ (0, 10, 100, 1000, 10,000 nM). After freezing-thawing, motility, viability, acrosome integrity, activity, adenosine triphosphate (ATP) levels, DNA malondialdehyde (MDA) reactive oxygen species (ROS) superoxide dismutase (SOD) transcription proteins fertilization capacity were assessed. The results show that 1000 nM supplementation significantly enhanced integrity compared control (p < 0.05). Additionally, increased membrane potential (MMP) ATP while decreasing MDA ROS reducing Furthermore, levels mitochondrial-encoded elevated group Interestingly, showed higher binding rate oviductal epithelial cells zona pellucida (ZP), indicating potential. These findings suggest use mitochondria-target PQQ, can improve via its reduce stress protect function.

Cryopreserved semen is extensively utilized in the artificial insemination (AI) of domestic animals; however, suboptimal conception rates due to oxidative damage following AI continue pose a challenge. The present study investigated effects Pyrroroquinoline Quinone (PQQ), novel antioxidant, on quality Holstein bulls during cryopreservation, as well its potential molecular mechanisms. Semen samples were diluted with varying concentrations PQQ (0, 50 μmol/L, 100 150 μmol/L) prior cryopreservation. Following freeze-thaw process, comprehensive evaluation was conducted assess sperm motility, plasma membrane integrity, acrosome and levels reactive oxygen species (ROS), malondialdehyde (MDA), adenosine triphosphate (ATP). Western blot analysis employed examine proteins including PGAM2, CAPZB, CAT, SOD1, GPX1. Notably, inclusion μmol/L significantly enhanced integrity post freeze-thawing (
