Abstract Rheumatoid arthritis (RA) is a systematic autoimmune disease that causes joint inflammation, pain and stiffness, leading to loss of function the joint. However, pharmacological treatments are usually accompanied by undesired adverse side effects, therefore, there has been relatively increased concern in other forms treatment. In this research, possible treatment effects Tinospora cordifolia examined given plant possesses anti-inflammatory immunomodulatory properties. Thus, in vivo vitro approaches were employed conducting research. In study entailed use extract on an animal model RA determine its therapeutic efficacy relation swelling, inflammation. Some animals received for pre – determined period clinical signs like paw swelling score assessed. The changes degree tissue injury inflammation at cellular level also assessed using histopathological examination. studies analyzed scientific performance immune cells secretion inflammatory cytokines. PBMCs prepared cultured with levels TNF-α IL-1β, IL-6 estimated. Cell viability regulation cytokine gene expression measured enzyme-linked immunosorbent assay (ELISA) quantitative real-time PCR, respectively. findings showed tested compounds reduced model, as well suppressed generation pro-inflammatory cytokines experiments. It further evidence may have activity could be used complementary or even alternative conventional medicines. more required before pinpointing bioactive accountable depicted herein.

Botanical extracts are recognized in traditional medicine for their therapeutic potential and safety standards. viable sustainable alternatives to synthetic drugs, being essential drug discovery various diseases. Senna septemtrionalis (Viv.) H.S. Irwin & Barneby is a medical plant traditionally used treat inflammation. However, its antioxidant anti-inflammatory properties the molecular pathways activated microglial cells require further investigation. Therefore, this study examines of methanol (SMEs) lipopolysaccharide (LPS)-stimulated mouse cells. SMEs significantly inhibit LPS-induced nitric oxide (NO) proinflammatory cytokine production, which mediated through dephosphorylation mitogen-activated protein kinases inhibition nuclear factor kappa B (NF-κB) translocation into nucleus. Additionally, SME treatment upregulated expression erythroid 2-related 2 (Nrf2) heme oxygenase (HO)-1, reducing oxidative stress, indicated by decrease reactive oxygen species restoration total glutathione content LPS-stimulated BV2 The inhibitory effects on inflammatory mediator production NF-κB were reversed Sn-protoporphyrin, specific HO-1 inhibitor. These findings demonstrate that protects activating Nrf2/HO-1 pathway inhibiting translocation.
