Oxidative stress is the key cause of etiopathogenesis several diseases associated with constipation. This study examined whether green pine cone can improve symptoms constipation based on antioxidant activities. The changes in parameters for activity and laxative effects were loperamide (Lop)-induced Sprague-Dawley (SD) rats after being treated methanol extracts (MPC, unripe fruits Pinus densiflora). MPC contained bioactive compounds, including diterpenoid compounds such as dehydroabietic acid, taxodone, ferruginol. In addition, it exhibited high scavenging against 2,2-diphenyl-1-picrylhydrazyl (DPPH) 2,2′-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radicals. These successfully reflected improvement nicotinamide adenine dinucleotide phosphate oxidase (NADP) H transcription, superoxide dismutase (SOD) levels, nuclear factor erythroid 2-related 2 (Nrf2) phosphorylation levels mid colon Lop+MPC-treated SD rats. Furthermore, significant improvements stool parameters, gastrointestinal (GI) transit, intestine length, histopathological structure detected Lop-induced treatment. other regulators adherens junction (AJ) tight (TJ), GI hormone secretion effects, improved significantly also verified primary rat smooth muscle cells (pRISMCs) through analyses defense mechanisms. Overall, finding this offers novel scientific evidence that could be considered a chronic its activity.

Colorectal cancer (CRC) is one of the most prevalent and deadly malignancies worldwide. Recently, ferroptosis, a novel form regulated cell death characterized by iron dependency lipid peroxidation, has garnered significant attention from researchers. The mechanisms underlying including intracellular levels, antioxidant system regulation, offer new insights into treatment strategies. This study aims to explore emerging role ferroptosis in context immunotherapy for CRC, highlighting its potential clinical applications. We employed comprehensive review current literature elucidate biological relationship with interplay between immunotherapy. Ferroptosis reshapes tumor microenvironment (TME) regulating metabolism, systems, significantly enhancing efficacy immune checkpoint inhibitors (ICIs). Meanwhile, traditional Chinese medicine therapies promote antitumor immunity modulating TME inducing ferroptosis. Additionally, advances nanotechnology have facilitated precise therapy enabling targeted delivery inducers or immunomodulators, transforming "cold" tumors "hot" further boosting ICI efficacy. comprehensively reviews latest developments immunotherapy, medicine, importance ferroptosis-related biomarkers personalized treatment. In summary, offers promising strategy overcome CRC resistance enhance efficacy, warranting investigation translational application.

Background: This study aims to reveal the potential molecular mechanisms of modified Gegen Qinlian decoction (MGQD) in relieving ulcerative colitis (UC). Methods: C57BL/6J mice were used establish experimental via dextran sodium sulfate (DSS). Body weight, disease activity index (DAI), spleen colon length, and histopathologic features measured evaluate therapeutic effects MGQD on with UC. The ELISA kits employed assess concentrations interleukin (IL)‐6, IL‐1β, tumor necrosis factor‐α (TNF‐α), glutathione (GSH), reactive oxygen species (ROS), malondialdehyde (MDA). Western blot analyses levels IκBα, p65, p‐IκBα, p‐p65, HO‐1, Nrf2. Moreover, protein Nrf2 p‐p65 assessed by immunofluorescence. Results: Colitis‐related symptoms significantly alleviated MGQD. inhibited TNF‐α, IL‐6, MDA, ROS increased level GSH Mechanistically, prevented activation NF‐κB pathway concomitantly promoted Nrf2/HO‐1 pathway. Conclusion: UC suppressing inflammation oxidative stress modulation pathways, suggesting that may be a candidate therapy for

Adequate protein consumption is essential for optimal physical fitness and enhancing athletic performance. This study explored the impact of Weizmannia coagulans BC99 on protein-supplemented male fatigued mice, examining aspects such as digestion, exercise endurance, fatigue-related biochemistry, oxidative stress, gut microbiota alterations. Results indicate that synergistic effect probiotics significantly boosts activity protein-digesting enzymes, enhances absorption, reduces serum levels urea nitrogen, lactate, lactate dehydrogenase, creatine kinase, malondialdehyde, inflammatory cytokines interleukin-1β interleukin-6 in skeletal muscle. Additionally, catalase, glutathione, superoxide dismutase levels, interleukin-4 muscle, glycogen stores muscle liver were notably increased. The also found elevated mRNA expression Nrf2 HO-1 Furthermore, an increase short-chain fatty acids was observed probiotic treatment group, 16S rDNA sequencing revealed enhanced diversity augmented beneficial bacterial populations including Roseburia, Mucispirillum, Rikenella, Kineothrix. Collectively, these findings suggest combining with supplementation can effectively improve flora, thereby capacity exerting potent anti-fatigue effects. Our research provides a new possibility alleviating exercise-induced fatigue.
