Immunoassays
based
on
the
specific
antigen-antibody
interactions
are
efficient
tools
to
detect
various
compounds
and
estimate
their
content.
Usually,
these
assays
implemented
in
water-saline
media
with
composition
close
physiological
conditions.
However,
many
substances
insoluble
or
cannot
be
molecularly
dispersed
such
media,
which
objectively
creates
problems
when
interacting
aquatic
environments.
Thus,
obtaining
immunoreactants
implementing
immunoassays
of
need
special
methodological
solutions.
Hydrophobicity
antigens
as
well
limited
ability
functionalization
conjugation
often
overlooked
developing
for
compounds.
The
main
key
finding
is
possibility
influence
behavior
hydrophobic
immunoassays,
requires
approaches
summarized
review.
Using
examples
two
groups
compounds-surfactants
(alkyl-
bisphenols)
fullerenes,
we
systematized
existing
knowledge
experience
development
immunoassays.
This
review
addresses
challenges
immunodetection
poorly
soluble
proposes
solutions
use
hydrotropes,
other
solubilization
techniques,
alternative
receptors
(aptamers
imprinted
polymers).

When
developing
immunochemical
test
systems,
it
is
necessary
to
obtain
specific
antibodies.
Their
quality
depends,
among
other
things,
on
the
immunogen
used.
preparing
hapten–protein
conjugates
antibodies
for
low-molecular-weight
compounds,
key
factors
are
structure
of
hapten
itself,
presence
a
spacer,
size
carrier
protein
and
degree
its
modification
by
molecules.
This
work
shows
that
one
additional
factor—the
conditions
obtaining
conjugate—is
overlooked.
In
this
work,
we
have
synthesized
bisphenol
A
derivative
4,4-bis(hydroxyphenyl)valeric
acid
(BVA),
soybean
trypsin
inhibitor
(STI),
bovine
serum
albumin
(BSA)
in
reaction
media
combining
water
with
two
organic
solvents:
dimethylformamide
(DMF)
or
dimethyl
sulfoxide
(DMSO).
Namely,
BSADMF–BVA,
STIDMF–BVA,
BSADMSO–BVA
STIDMSO–BVA
were
obtained.
Rabbit
polyclonal
against
BSADMF–BVA
conjugate
demonstrated
basically
different
interactions
developed
ELISA
systems
using
either
STIDMF–BVA
conjugates.
The
use
absence
competition
combination
antisera
obtained
from
an
ELISA.
competitive
interaction
was
observed
only
conjugate.
Under
selected
conditions,
detection
limit
8.3
ng/mL,
working
range
determined
concentrations
18.5–290.3
ng/mL.
data
demonstrate
possibility
achieving
sensitive
immunoassays
simply
varying
conjugation,
which
could
provide
tool
development
compounds.
