Abstract Objectives Bladder cancer (BLCA) is a tumor that affects men more than women. The biological function and prognostic value of androgen-responsive genes (ARGs) in BLCA are currently unknown. To address this, we established an androgen signature to determine the prognosis BLCA. Methods Sequencing data for from TCGA GEO datasets were used research. microenvironment (TME) was measured using Cibersort ssGSEA. Prognosis-related identified risk score model constructed univariate Cox regression, LASSO multivariate regression. Drug sensitivity analysis performed Genomics drug (GDSC). Real-time quantitative PCR assess expression representative clinical samples. Results ARGs (especially CDK6, FADS1, PGM3, SCD, PTK2B, TPD52) might regulate progression different patterns lead immune cell infiltration. indicates patients with higher scores have poorer prognosis, stromal infiltration, enrichment functions. Single-cell RNA analysis, bulk data, proved reliability this model, nomogram also use. prediction showed high-risk had better response fludarabine, AZD8186, carmustine. Conclusion played important role progression, has high accuracy predicting provides effective medication guidelines.

Circular RNA (circRNAs) is a covalently closed circular non-coding formed by reverse back-splicing from precursor messenger RNA. It found widely in eukaryotic cells and can be released to the surrounding environment captured other cell types. This, circRNAs serve as connections between different types for mediation of multiple signaling pathways. CircRNAs reshape tumor microenvironment (TME), key factor involved all stages cancer development, regulating epithelial-stromal transformation, vascularization, immune function, inflammatory responses. Immune are most abundant cellular TME components, they have profound toxicity cells. This review summarizes circRNA regulation cells, including T natural killer macrophages; highlights impact on progression, treatment, prognosis; indicates new targets immunotherapy.

Abstract The bladder and prostate originate from the urogenital sinus. However, cancer (BC) is usually classified as an immune “hot” tumor, whereas (PCa) deemed “cold” tumor according to microenvironment (TME) clinical outcomes. To investigate differences between BC PCa, studies are compared focusing on regulation mediated by sex hormones receptors identify key genes pathways responsible for differences. From a developmental perspective, it shown that PCa activate similar those in stage. During development, differential expression function of androgen receptor (AR) across cell types may contribute its dual role promoting inhibiting immunity different cells. Androgen deprivation therapy affects AR cells within TME, influencing infiltration antitumor function. Additionally, estrogenα estrogenβ exert contrasting effects BC, which hold potential modifying phenotypes. Future research should target involved development clarify regulatory similarities PCa.

Males have a three times higher risk of diagnosis bladder cancer (Bca) than females. Sex hormone-binding globulin (SHBG) may be associated with Bca risk. However, the sex-specific role SHBG in remains unclear. In this study, we aimed to determine Bca. A univariable Mendelian randomization (MR) analysis involving 369,426 men and 395,375 women was conducted assess causal relationship between testosterone Sensitivity analyses multivariable MR were confirm robustness our results. Linkage disequilibrium score regression assessed genetic correlation these diseases influenced by heredity. Univariable results showed that one-SD elevated related low males (OR: 0.60, 95% CI: 0.39-0.93; p = 0.022) but had no benefit Genetically predicted BT positively 1.59; 1.06-2.40; 0.027).
