Crohn's disease (CD) is a chronic inflammatory autoimmune of unknown etiology. To identify new targets related to the initiation CD, we screened pair twins with which rare phenomenon in Chinese population, for genetic susceptibility factors. Whole-exome sequencing (WES) these patients revealed mutation their SERPINB4 gene. Therefore, studied wider clinical cohort CD or ulcerous colitis (UC), healthy individuals, and those family history this by Sanger sequencing. The single-nucleotide difference gene, was unique twin led substitution lysine glutamic acid residue. Functional analysis indicated that inhibited proliferation, colony formation, wound healing, migration intestinal epithelial cells (IECs). Furthermore, induced apoptosis activated apoptosis-related proteins IECs, caspase inhibitor significantly reduced effects. Transcriptome expression genes encoding proinflammatory (IL1B, IL6, IL17, IL24, CCL2, CXCR2) key immune response (S100A9, MMP3, MYC) upregulated during mutant-induced apoptosis. Thus, heterozygous gene causes dysfunction would disrupt barrier contribute development inflammation. activation might represent novel therapeutic target bowel disease.

Laminin subunit alpha-5 (LAMA5) has been identified as an oncogene in many cancers, while its role and mechanism gastric cancer (GC) remain to be explored. Here, the influences of LAMA5 knockdown on GC were investigated vitro vivo. expression was silenced cells alone or combination with signal transducer activator transcription 3 (STAT3) Colivelin, followed by CCK-8, colony formation, EdU, flow cytometry, wound healing assay, Transwell assay. The regulatory relationship between Myc-associated zinc finger protein (MAZ) characterized ChIP luciferase reporter analysis. effect MAZ overexpression highly expressed cells, inhibited cell malignant aggressiveness, which reversed Colivelin treatment. factor bound promoter activate transcription, anti-tumor effects sh-MAZ vivo overturned overexpression. In conclusion, promotes proliferation migration LAMA5/STAT3 axis, implying that this axis can function a target for therapy.

Scutellarin (SC), a natural flavonoid, has been expansively employed in treating innumerable inflammation-related diseases due to its antitumor, antiinflammatory, anticancer, and antioxidant potential. can inhibit significant inflammatory cell signaling pathways, comprisingPI3K/Akt, NF-κB, MAPK, while activating antioxidant-related pathways such as Nrf2 ARE. Numerous reviews have outlined scutellarin’s pharmacological effects associated mechanisms diseases. Several studies elucidated the of anticancer activity by inhibiting various pathways; however, our knowledge, none distinguished potential scutellarin based on different human cancer types. Our review detailed insights about type body. Furthermore, we also formulations, combinatorial therapies, comprehensive mechanistic research deliver enhanced effective treatment options for patients. This study will provide thorough into scutellarin, supporting development promising candidate treatment.

ABSTRACT Signal transducer and activator of transcription 3 (STAT3) is a critical factor involved in multiple physiological pathological processes. While STAT3 plays an essential role homeostasis, its persistent activation has been implicated the pathogenesis various diseases, particularly cancer, bone‐related autoimmune disorders, inflammatory cardiovascular neurodegenerative conditions. The interleukin‐6/Janus kinase (JAK)/STAT3 signaling axis central to activation, influencing tumor microenvironment remodeling, angiogenesis, immune evasion, therapy resistance. Despite extensive research, precise mechanisms underlying dysregulated disease progression remain incompletely understood, no United States Food Drug Administration (USFDA)‐approved direct inhibitors currently exist. This review provides comprehensive evaluation STAT3's health disease, emphasizing involvement cancer stem cell maintenance, metastasis, inflammation, drug We systematically discuss therapeutic strategies, including JAK (tofacitinib, ruxolitinib), Src Homology 2 domain (S3I‐201, STATTIC), antisense oligonucleotides (AZD9150), nanomedicine‐based delivery systems, which enhance specificity bioavailability while reducing toxicity. By integrating molecular mechanisms, pathology, emerging interventions, this fills knowledge gap STAT3‐targeted therapy. Our insights into crosstalk, epigenetic regulation, resistance offer foundation for developing next‐generation with greater clinical efficacy translational potential.

Salidroside (SAL) is a bioactive substance extracted from the traditional Chinese medicine Rhodiola rosea, which exhibits multiple pharmacological effects, such as anti-inflammatory, antioxidant, and anti-tumor properties. Currently, effects of SAL on malignant progression ovarian cancer (OC) its specific mechanism action are not clear. Cell Counting Kit 8 (CCK-8), clone formation, Hoechst 33258 staining, flow cytometry, transwell, western blotting immunofluorescence assays were performed to determine impacts biological properties OC cells (CAOV3 SKOV3) human normal epithelial (IOSE80). The binding activity proteins was evaluated. Glucose consumption, lactate ATP production, extracellular acidification rate (ECAR) related measured assess glycolysis. Animal models established evaluate impact treatment in vivo expression levels STAT3/c-Myc pathway-related determined explore relationship between OC. results showed that reduced viability, migration invasion ability CAOV3 SKOV3 cells, induced apoptosis. inhibited epithelial-mesenchymal transition (EMT) decreased glucose production ECAR. exhibited good with STAT3 c-Myc pathway glycolysis-related vitro vivo.
