One-carbon (1C) metabolism is a complex network of metabolic reactions closely related to producing 1C units (as methyl groups) and utilizing them for different anabolic processes, including nucleotide synthesis, methylation, protein reductive metabolism. These pathways support the high proliferative rate cancer cells. While drugs that target (like methotrexate) have been used treatment, they often significant side effects. Therefore, developing new with minimal effects necessary effective treatment. Methionine, glycine, serine are main three precursors vital not only cells but also non-proliferative in regulating energy homeostasis aging process. Understanding potential role crucial advancing our knowledge neoplastic progression. This review provides comprehensive understanding molecular complexities context aging, paving way researchers explore avenues advanced therapeutic interventions cancer.

Introduction and Objective: Obesity has increased worldwide, existing anti-obesity medications have treatment limitations that diminish their overall benefits. This study aimed to investigate the effects of orlistat in combination with Elettaria cardamomum “Cardamom” (CAR) extract on working memory, recognition anxiety, inflammation within hippocampal tissue. Methods: Mice were categorized into two groups: a control group (CD) cafeteria diet (CAF) induced obesity for 10 weeks. The groups then subdivided CAF treated (CAF-ORL), (CAF-ORL-CARD), continued CAF. CAF-ORL received at dosage mg/kg/day four weeks, while CAF-ORL-CARD 500 mg/kg CAR via oral gavage. In 14th week, various assessments conducted, including novel object (NOR) test, Y maze marble-burying test (MBT), open-field TNF-α levels hippocampus. Result: tissue elevated compared CD (p < 0.01), whereas exhibited reduced 0.01). Moreover, significantly lower than index was notably higher 0.01) However, there no changes > 0.05). Conclusions: Orlistat combined positive neuroinflammation suggesting this may offer potential therapeutic benefits cognitive impairments dysfunction associated obesity.

Abstract Sono-photodynamic therapy (SPDT) has attracted a lot of interest as cutting-edge therapeutic strategy in the field cancer treatment. The essential part SPDT is sensitizer, which under laser photon and pulsed cavitation ultrasound sono-irradiation may transform sono photo- energy into cytotoxic molecules. Photon absorption, targeting, penetration, oxygen dependence remain challenges sono- -photosensitizer (SPs) design. Rapid advancements material science have prompted creation several SPs that create species with great selectivity, safety, noninvasiveness for treatment tumors. current study aims to provide an advanced method activated by using assist delivery cardamom, pistacia laurel conjugated micelles nanoparticles (CPL-Micelle NP) sono-photodynamic lymphoma vivo vitro Human cells (U-937) were used study, application groups protocol Swiss albino mice treated 9,10-Dimethyl-1,2-Benzanthracene (DMBA) only; they not given any induce lymphoma. started only after induction, involved daily administration CPL-Micelle NP sensitizer whether or be exposed (IRL) (US) combination them three minutes period two weeks. Indicated Micelle useful CPL mechanism targets directly. Furthermore, promising SPS that, when conjunction SPDT, can very effective treating lymphoma-U-937 (in dose-dependent manner cell viability declined, increase population during S G2/M phases indicates cycle was arrested, Pre-G, autophagic death, well necrosis early late apoptosis, indicate death induced) DMBA-Lymphoma-induced (induced antiproliferative genes, repressed antiangiogenic antiapoptotic genes), successfully slowing growth tumors even killing cells, lowering oxidative stress malondialdehyde (MDA), improving functions kidneys, liver, enzymatic non antioxidants. chemical activation mechanism, antioxidant capacity non-activated all linked this process. On bases findings, shows promise novel, efficient selective system localized SPDT-activated

Metabolic reprogramming is a hallmark of cancer and de novo lipogenesis (DNL) accelerates the progression ovarian cancer. In this study, we investigated effects cardamonin, natural compound potential to suppress various malignancies, on lipid anabolism in Cell proliferation was assessed using CCK-8 clone formation assay. apoptosis detected by flow cytometry with Annexin V-FITC/PI staining mitochondrial membrane (MMP) measured JC-10 probe. Free fatty acids (FFA) fluorescence acyl-CoA oxidation carnitine palmitoyl transferase-1 (CPT-1) activity analyzed spectrophotometric assay palmitoyl-CoA DTNB (5,5’-dithio-bis-(2-nitrobenzoic acid)) reaction. mRNA expression Quantitative Real-Time PCR. Protein through western blotting immunofluorescence. Raptor knocked down shRNA overexpressed lentiviral transfection. The antitumor effect cardamonin evaluated xenotransplantation tumor bearing mouse model. Cardamonin suppressed cell proliferation, induced triggered damage cells. inhibited protein sterol regulatory element binding 1 (SREBP1) its downstream lipogenic enzymes decreased FFA content CPT-1 activity. Additionally, activation mechanistic target rapamycin complex (mTORC1) regulatory-associated mTOR (Raptor). knockdown abolished inhibitory mTORC1 SREBP1. Furthermore, proteins overexpression. reduced growth acid synthase tumors, as evidenced Ki-67 FASN. It suggests that suppresses mTORC1/SREBP1 reducing level inhibits DNL
