Epimedium has been widely used in traditional Chinese medicine for several thousands of years. This plant is known tonifying kidney Yang, strengthening muscles and bones, dispelling wind dampness. It worth noting that icaritin, a prenylated flavonoid isolated from Epimedium, received increasing attention recent years due to its wide range pharmacological activities. Icaritin exhibits significant therapeutic potential against various diseases, such as osteoporosis, tumors (hepatocellular carcinoma, stomach cancer, breast glioblastoma), cerebral ischemia skin injury, thrombocytopenia, systemic lupus erythematosus. We review the activities icaritin molecular mechanisms treatment related diseases. The data suggest can have effects mediating Wnt/[Formula: see text]-catenin, IL-6/JAK2/STAT3, AMPK/mTOR, PTEN/AKT, MAPK, NF-[Formula: text]B, other signaling pathways. paper also discusses progress clinical trials icaritin. was approved by State Food Drug Administration January 2022 advanced HCC, drug prospects. Although it some disadvantages, including poor solubility, low bioavailability, still prospective candidate development naturally derived drugs, especially inflammatory aims update deepen understanding provide theoretical basis further study.
