Lung cancer is one of the leading causes death worldwide and most common all types. Histone acetyltransferase 1 (HAT1) has attracted increasing interest as a potential therapeutic target due to its involvement in multiple pathologies, including cancer. Aptamers are single-stranded RNA or DNA molecules whose three-dimensional structure allows them bind molecule with high specificity affinity, thus making exceptional candidates for use diagnostic tools. In this work, aptamers against HAT1 were obtained, subsequently characterized, optimized, showing affinity ability inhibit activity vitro. Of those tested, apHAT610 aptamer reduced cell viability, induced apoptosis cycle arrest, inhibited colony formation lung lines. All these results indicate that drug treatment

Background & Objectives: Systematic evolution of ligands by exponential enrichment (SELEX) technology was widely used to screen the aptamers that bind target protein safely and efficiently. Our study aimed for anti-depression via binding 5-HTR block 5-HT re- uptake. Methods: The prokaryotic expression plasmid constructed recombinant 5-HT1AR (mice) expressed purified. ssDNA aptamer bound specifically screened SELEX (Enzyme-linked Oligonucleotide assay), sites relative strength were detected. At same time, inhibitory uptake against depression verified in cellular level mouse model. Results: purified successfully. After 12 rounds positive screening 5 negative screening, four with high affinity specificity obtained epitope bounded using ELONA assay). influenced 18 vitro, improvement state mice after intravenous injection proved tail suspension experiment mice. Conclusions: Aptamer is expected be a new type antidepressant, which can treatment depression.
