Abstract Among the most important agro‐industrial activities in Mediterranean basin, olive oil production has a high impact on economy of many countries. However, extraction generates huge quantities byproducts, including leaves, pomace residues, stones and wastewater, which have severe environmental impacts mainly because their phytotoxicity great organic content. Olive byproducts are regarded as inexpensive abundant raw materials rich bioactive compounds with varied health‐related activities. Several phenolic terpenoids were recovered from using different conventional advanced methods due to potential be used food, packaging, pharmaceutical, cosmetic industries. Recently, use functional enhance properties packaging systems was investigated sustainable strategy for food preservation, fostering sustainability olive‐oil chain, promoting circular economy. Olive oil contains different biologically active polyphenols, among which oleacein, the most abundant secoiridoid, has recently emerged for its beneficial properties in various disease contexts. By using vitro models of human multiple myeloma (MM), we here investigated anti-tumor potential oleacein and underlying bio-molecular sequelae. Within a low micromolar range, reduced viability MM primary samples cell lines even presence bone marrow stromal cells (BMSCs), while sparing healthy peripheral blood mononuclear cells. We also demonstrated that inhibited clonogenicity, prompted cycle blockade triggered apoptosis. evaluated epigenetic impact on cells, observed dose-dependent accumulation both acetylated histones α-tubulin, along with down-regulation several class I/II histone deacetylases (HDACs) at mRNA protein level, providing evidence HDAC inhibitory activity this compound; conversely, no effect global DNA methylation was found. Mechanistically, HDACs inhibition by associated Sp1, major transactivator promoter, via Caspase 8 activation. Of translational significance, synergistically enhanced anti-MM proteasome inhibitor carfilzomib. Oleuropein plays a key role as pro-oxidant well an antioxidant in cancer. In this study, the activity of oleuropein, vitro model ovarian (OCCs) and breast cancer cells (BCCs) was investigated. Cell viability cell death were analyzed. Oxidative stress measured by CM-H2DCFDA flow cytometry assay. Mitochondrial dysfunction evaluated based on mitochondrial reactive oxygen species (ROS) GPX4 protein levels. Further, effects iron metabolism analyzed measuring intracellular labile pool (LIP). We confirmed that high doses oleuropein show anti-proliferative pro-apoptotic HEY MCF-7 cells. Moreover, our results indicate low impair without affecting mortality cells, also decrease LIP ROS levels, keeping them unchanged For first time, data reduce erastin-mediated death. Interestingly, decreases levels OCCs treated with erastin. Noteworthily, we observed increased amount scavenging enzyme together consistent reduction ROS, confirming oxidative model.

Breast and ovarian cancers are the leading fifth reason for tumor death among females, respectively. Recently, many studies demonstrated antiproliferative activities of natural aliments in cancer. In this study, we investigated antitumor potential Olive Leaf Extract (OLE) triple-negative breast cancer cells. A HPLC/DAD analysis on OLE has been performed to assess total polyphenolics other secondary metabolites content. HCEpiC, MDA-MB-231, OVCAR-3 cell lines were used. MTS, Cytofluorimetric, Western Blot analyze viability, proliferation, apoptosis, oxidative stress. Fluorimetric IncuCyte® analyses carried out evaluate apoptosis mitochondrial function. We confirmed that OLE, containing a quantity oleuropein 87 % extract, shows anti-proliferative pro-apoptotic activity MDA-MB-231 For first time, our results indicate inhibits viability inducing cycle arrest, it also increases apoptotic up-regulating protein level cleaved-PARP caspase 9. Moreover, data show treatment causes significant decrease functionality, paralleled by reduction membrane potential. Interestingly, increased intracellular reactive oxygen species (ROS) together with decreased ROS scavenging enzymes, confirming stress both models. Our demonstrate generation represented primary mechanism activity, as pretreatment antioxidant N-acetylcysteine prevented OLE-induced arrest apoptosis.
