Abstract 5- methylcytosine (m5C) is a prevalent posttranscriptional RNA modification which participates in the initiation and progression of various cancers. NSUN2 main methyltransferase import for catalyzing m5C formation. However, its regulatory role potential mechanism osteosarcoma remain unclear. Here, we demonstrated that expression was markedly upregulated tissues cell lines. Clinically, increased associated with poor prognosis. Functional studies revealed significantly promoted metastasis epithelial-mesenchymal transition (EMT) osteosarcoma. Mechanistically, integrated analysis based on sequencing correlation identified UBE2S as target downstream gene NSUN2, while enhanced mRNA stabilization an m5C-dependent manner. More importantly, UBE2S overexpression reversed inhibition invasion EMT induced by knockdown. Moreover, interacted ubiquitinated β-catenin, enhancing stability activation. Interestingly, patients dual-high exhibited shorter overall survival. In summary, our study facilitated UBE2S/β-catenin axis, suggesting therapeutic approach
