We have recently developed a machine learning classifier that enables fast, accurate, and affordable classification of brain tumors based on genome-wide DNA methylation profiles is widely employed in the clinic. Neuro-oncology research would benefit greatly from understanding underlying artificial intelligence decision process, which currently remains unclear. Here, we describe an interpretable framework to explain classifier's decisions. show functional genomic regions various sizes are predominantly distinguish between different tumor classes, ranging enhancers CpG islands large-scale heterochromatic domains. detect high degree redundancy, with many genes distinguishing individual explaining robustness revealing potential targets for further therapeutic investigation. anticipate our resource will build up trust clinical settings, foster biomarker discovery development compact point-of-care assays, enable epigenome tumors. The pituitary is the central endocrine gland with effects on metabolic dysfunction-associated steatotic liver disease (MASLD). However, it not clear whether responds to free fatty acid (FFA) toxicity, thus dysregulating hepatic lipid metabolism. Here, we demonstrate that decreased prolactin (PRL) levels are involved in association between FFA and MASLD based a biospecimen-based cohort. Moreover, overloaded FFAs decrease serum PRL levels, promoting steatosis mice both dynamic diet intervention stereotactic injection. Mechanistic studies show excessive sensing lactotrophs inhibits synthesis secretion of cell-autonomous manner. Notably, inhibiting uptake using stereotaxic virus injection or specific drug delivery system effectively ameliorates accumulation by improving levels. Targeted inhibition may be potential therapeutic target for steatosis.

Pituitary neuroendocrine tumors (PitNET) are among the most common intracranial tumors. Despite a frequent benign course, aggressive behavior can occur. Tumor is known to be under influence of tumor microenvironment (TME). However, relationship between TME cells and has not been adequately explored in PitNET. We performed differential expression analysis as well gene program identification based on single-cell RNA sequencing comparatively characterize transcriptome seven gonadotroph three lactotroph PitNET correlate it with clinical features using bulk RNA-seq data from an independent cohort 134 immune infiltration was quantified via immunostaining tissue sections In PitNET, we detect highly proliferative profile significantly increased levels aggressively growing within samples. also report high intratumoral heterogeneity (GoPN) (LaPN) identify signatures epithelial, endocrine, immunological networks both subtypes. A comparison their composition shows enrichment SPP1+ macrophages CD4+ T GoPN, CD4/CD8 double-negative (DN) natural killer (NK) LaPN. Also notable presence lymphocytes, occurrence which positively correlates more cohort. CD8+ NK cell abundances reduced aggressiveness indicating potential anti-tumoral effects. Our study expands knowledge differences cellular It lays foundation for further studies lymphoid variable Regarding treatment drug-resistant T, could represent potentially valuable targets developing new cancer immunotherapies.
