Background: Neurofibromin, coded by the NF1 tumor suppressor gene, is main negative regulator of RAS pathway and frequently mutated in various cancers. Women with Neurofibromatosis Type I (NF1)–a predisposition syndrome caused a germline mutation–have an increased risk developing aggressive breast cancer poorer prognosis. The mechanism which mutations lead to tumorigenesis not well understood. Therefore, objective this work was identify stromal alterations before formation that result outcome seen among patients cancer. Approach: To accurately model monoallelic patients, we utilized Nf1- deficient rat accelerated mammary development presenting highly penetrant Results: We identified collagen content Nf1 -deficient glands correlated age onset. Additionally, gene expression analysis revealed mature adipocytes gland have shifted fibroblast preadipocyte profile. This alteration lineage commitment also observed vitro differentiation, however, flow cytometry did show change adipose-derived mesenchymal stem cell abundance. Conclusion: Collectively, study uncovered previously undescribed role deposition regulating adipocyte differentiation. In addition unraveling formation, further investigation modifications preneoplastic will create foundation for early detection strategies patients.

Breast cancer has been reported to transcend lung as the most commonly diagnosed in women all over world. Adipocytes, serving energy storage and endocrine cells, are major stromal cells breast. Cancer-associated adipocytes (CAAs) adjacent dedifferentiated located at invasive front of human breast tumors. Adipocytes can transform into CAA phenotype with morphological biological changes under remodeling cells. CAAs play an essential role progression, including tumor microenvironment (TME), regulating immunity, interacting possess peculiar secretomes accordingly capable promote proliferation, invasiveness, angiogenesis, metastasis, immune escape, drug resistance There is a complex coordinated crosstalk among CAAs, release variety cytokines, IL-6, IL-8, IL-1β, CCL5, CCL2, VEGF, G-CSF, IGF-1, IGFBP, thereby promoting cell recruitment macrophage polarization, ultimately stimulating malignant behaviors Here, we aim provide comprehensive description CAA-derived their impact on behaviors, regulation, diagnosis, treatment. A deeper understanding performance interactions specific TME populations will better strategies for treatment reconstruction after mastectomy.

This study investigated whether the textural features of peritumoral adipose tissue (AT) on [18F]fluoro-2-deoxy-2-d-glucose (FDG) positron emission tomography/computed tomography (PET/CT) can predict pathological response to neoadjuvant chemotherapy (NAC) and progression-free survival (PFS) in breast cancer patients. We retrospectively enrolled 147 female patients who underwent staging FDG PET/CT completed NAC curative surgery. extracted 10 first-order features, 6 gray-level co-occurrence matrix (GLCM) 3 neighborhood difference (NGLDM) AT evaluated predictive value those imaging for complete (pCR) PFS. The results our demonstrated that GLCM homogeneity showed highest predictability pCR among receiver operating characteristic curve analysis. In multivariate logistic regression analysis, mean standardized uptake (SUV), 50th percentile SUV, 75th SUV histogram entropy, were independent predictors pCR. entropy correlation Textural could be potential biomarkers predicting disease progression

Cancer cells-derived extracellular vesicles can trigger the transformation of adipose-derived mesenchymal stem cells (ADMSC) into a pro-inflammatory, cancer-associated adipocyte (CAA) phenotype. Such secretome-mediated crosstalk between adipose tissue and tumor microenvironment (TME) therefore impacts progression metastatic processes. In addition, emerging roles diet-derived phytochemicals, especially epigallocatechin-3-gallate (EGCG) among other polyphenols, in modulating exosome-mediated metabolic inflammatory signaling pathways have been highlighted. Here, we discuss how selected phytochemicals could alter secretome signature as well dynamics TME, with focus on breast cancer. Their broader implication chemoprevention obesity-related cancers is also discussed.
