Abstract Mitochondrial dysregulation plays a significant role in the carcinogenesis. On other hand, its destabilization strongly represses viability and metastatic potential of cancer cells. Photodynamic photothermal therapies (PDT PTT) target mitochondria effectively, providing innovative non-invasive anticancer therapeutic modalities. Cyanine dyes, with strong mitochondrial selectivity, show enhancing PDT PTT. The limitations cyanine dyes for PTT are discussed, along their applications combination therapies, theranostic techniques, optimal delivery systems. Additionally, novel approaches sonodynamic therapy using photoactive presented, highlighting advances treatment.

Triple-negative breast cancer (TNBC) is a subtype of that presents significant therapeutic challenges due to the absence estrogen receptor (ER), progesterone (PR), and human epidermal growth factor 2 (HER2) expression. As result, conventional hormonal targeted therapies are largely ineffective, underscoring urgent need for novel treatment strategies. γδT cells, known their robust anti-tumor properties, show considerable potential in TNBC as they can identify eliminate tumor cells without reliance on MHC restrictions. These demonstrate extensive proliferation both vitro vivo , directly target tumors through cytotoxic effects or indirectly by promoting other immune responses. Studies suggest expansion adoptive transfer strategies targeting Vδ2 Vδ1 cell subtypes have shown promise preclinical models. The persisting presence of opportunistic pathogens like Pseudomonas aeruginosa poses a significant threat to many immunocompromised cancer patients with pulmonary infections. This review highlights the complexity interactions in host's defensive eicosanoid signaling network and its hijacking by pathogenic bacteria their own advantage. Human lipoxygenases (ALOXs) mouse counterparts are integral elements innate immune system, mostly operating pro-inflammatory mode. Taking into account indispensable role inflammation carcinogenesis, have counteracting roles this process. In addition describing structure-function review, we discuss such critical processes as cell signaling, metastases, death cells through ferroptosis, well ALOXs carcinogenesis promoted Finally, perspectives novel oncotherapeutic approaches harness lipoxygenase tumors.

Breast cancer is the most prevalent among women in United States, representing ~30% of all new female cases annually. For year 2024, it estimated that 310,720 instances invasive breast will be diagnosed, and responsible for over 42,000 deaths women. Today, despite availability numerous treatments its symptoms, cancer-related result from metastasis which there no treatment. This emphasizes importance early detection treatment before spreads. initial staging cancer, clinicians routinely employ mammography ultrasonography, which, while effective broad screening, have limitations sensitivity specificity. Advanced biomarkers could significantly enhance precision detection, enable more accurate monitoring disease evolution, facilitate development personalized plans tailored to specific molecular profile each tumor. would not only improve therapeutic outcomes, but also help avoiding overtreatment associated side effects, thereby improving quality life patients. Published: Dec. 2, 2024

Microsomal prostaglandin E2 synthase 1 (mPGES-1) is a promising target for treating inflammatory diseases and pain. This study introduces novel series of benzimidazoles, with the most potent analogs exhibiting IC50 values 0.27–7.0 nM in cell-free assay (PG)E2 production. Compound 44 (AGU654) demonstrated remarkable selectivity mPGES-1 (IC50 = 2.9 nM) over COX-1, COX-2, 5-LOX, FLAP, along excellent bioavailability. Metabololipidomics analysis activated human monocyte-derived macrophages whole blood revealed that AGU654 selectively suppresses PGE2 production triggered by bacterial exotoxins while sparing other prostaglandins. Furthermore, vivo studies showed significantly alleviated fever, inflammation, pain preclinical guinea pig models, suggesting it could be an effective strategy managing diseases. In conclusion, these benzimidazole derivatives warrant further exploration into new alternative analogs, potentially uncovering compounds favorable pharmacological profile possessing significant anti-inflammatory analgesic properties.

Objective This study aims to identify safety signals of ophthalmic prostaglandin analogues through data mining the Food and Drug Administration Adverse Event Reporting System (FAERS) database. Methods A search by proportional reporting ratio, OR, Bayesian confidence propagation neural network, information component 0.25 χ 2 for detection was conducted FAERS database following medications: latanoprost, travoprost, tafluprost bimatoprost. Results 12 preferred terms were statistically associated: diabetes mellitus, n=2; hypoacusis, malignant mediastinal neoplasm, n=1; blood immunoglobulin E increased, cataract, blepharospasm, full count abnormal, skin exfoliation, chest discomfort, dry mouth, n=1. Limitation The database’s limitations, such as undetermined causality cases, under-reporting lack restriction only health professionals this type event, could modify statistical outcomes. These limitations are particularly relevant in context drug analysis, they can affect accuracy reliability data, potentially leading biased or incomplete results. Conclusions Our findings have revealed a potential relationship due biological plausibility among cataracts, discomfort mouth; therefore, it is continue investigating possible drug-event association, whether refute signal new risk.
