Ferroptosis has been recognized as a critical factor in the development of atrial fibrillation (AF), but its precise mechanisms remain unclear. We downloaded GSE115574 dataset from gene expression omnibus database to analyze levels ferroptosis-related genes (FRGs) and identify differentially expressed (DEGs). Least absolute shrinkage selection operator (LASSO) support vector machine-recursive feature elimination (SVM-RFE) machine learning techniques were employed key associated with AF. The diagnostic performance these was evaluated using Receiver operating characteristic curves (ROC) validated an independent AF dataset. miRNA lncRNA predictions for potential binding conducted miRBase, miRDB, TargetScan. Furthermore, set enrichment analysis (GSEA) analysis, immune cell infiltration targeted drug prediction performed. intersection LASSO regression SVM-RFE analyses identified 7 DEGs significantly Validation through ROC additional confirmed importance MAPK14, CAV1, ADAM23. Significant memory B cells, regulatory T monocytes observed tissues. Seventy-two miRNAs predicted potentially target 2 drugs targeting CAV1. This study underscores involvement FRGs validation approaches. suggests link between response ceRNA network offers new insights into regulation, presenting biomarkers therapeutic targets

Abstract Atrial fibrillation (AF) is a common arrhythmia associated with severe outcomes like heart failure and stroke. Recent studies highlight the crucial role of T in AF. Clinical have observed elevated levels CD4 + CD28 null cells, Th17/Treg CD8 related markers peripheral blood or atrial tissue AF patients, correlating disease severity cardiovascular events. These cell subsets contribute to through: (1) releasing inflammatory factors TNF-α IL-17 which affect calcium homeostasis electrical activity myocytes and/or promote fibrosis; (2) recruiting cells such as macrophages, causing local inflammation, oxidative stress, remodeling; (3) secreting cytotoxic proteins perforin granzymes, inducing apoptosis affecting their action potentials; (4) direct contact, influencing myocyte electrophysiology. Understanding these cell-mediated mechanisms may uncover new therapeutic targets for

Background The association between the Neutrophil-to-Lymphocyte Ratio (NLR) and prognosis of Atrial Fibrillation (AF) has been extensively studied, yet clinical outcomes have varied. Consequently, this analysis was undertaken to explore link NLR prognostic markers AF. Methods We conducted an exhaustive search across electronic databases, including PubMed, Embase, Web Science, Cochrane Library, investigate correlation indicators adverse associated with AF from database establishment date through March 31, 2024. In study, recurrence rate primary outcome measure, while secondary measures were mortality, stroke, left atrial thrombus. Odds ratio (OR), relative risk (RR), hazard (HR) standard mean difference (SMD) a 95% confidence interval (CI) integrated for assessment, stability publication bias verified by sensitivity Egger's test, respectively. Subgroup analyses performed pinpoint sources heterogeneity. Results This included 20 studies, encompassing total 59,256 patients. Our statistical both categorical continuous variables revealed that elevated significantly increased risks in patients (categorical variable: OR = 1.39, CI 1.21–1.60; SMD 0.49, 0.24–0.74), mortality 1.87, 1.59–2.20), stroke 1.56, 1.13–2.17; 0.77, 0.63–0.91), thrombus 1.27–2.75; 0.59, 0.30–0.89). found high linked when >3. High ≤3. Conclusions study suggested is AF, may be effective biomarker practice. Systematic Review Registration PROSPERO (CRD42024530970).
