Pseudogalium is a new monotypic genus with two subspecies in China and one Japan, which holds distinctive phylogenetic position ecological significance within the tribe Rubieae. Chloroplast genomes contain abundant information for resolving relationships. To investigate phylogenetics of P. paradoxum its related genera, we first sequenced, assembled, annotated chloroplast genome reconstructed trees. Due to lack samples subsp. franchetianum from this study only analyzed discussed duthiei. This had shown that complete ranged 153,093 bp 153,333 length 130 genes total, all typical circular structures consisting large single-copy region, small region pair inverted repeat regions. The comparative analysis showed was conserved Additionally, identified 60 dispersed sequences, 61-63 simple sequence repeats, 30 codons 82 protein-coding exhibited RSCU values greater than one. Furthermore, detected highly divergent regions hold potential as DNA barcodes species identification. Compared Pseudogalium, gene number, length, GC content Galium Rubia differential characteristics, sequences SSRs were significantly different. Phylogenetic based on whole can be treated genus, duthiei considered distinct paradoxum. reported study, provided insight into phylogeny taxonomy genus. analyses strongly supported following proposals: (1) isolated closely Galium; (2) form clades, both
