Although an association between metabolic dysfunction-associated fatty liver disease (MAFLD) and cardiovascular or overall mortality has been reported, it is unclear whether there MAFLD cancer incidence mortality. We aimed to investigate the differential risk of all- site-specific according subgroups categorized by additional etiologies disease.Using Korean National Health Insurance Service database, we stratified participants into three groups: (1) single-etiology (S-MAFLD) pure origin; (2) mixed-etiology (M-MAFLD) with etiological factor(s) (i.e., concomitant diseases and/or heavy alcohol consumption); (3) non-MAFLD. Hepatic steatosis fibrosis were defined using index BARD score, respectively. Obesity is a cancer progression risk factor; excessive adipocytes increase adipokine secretion. Visfatin, novel highly expressed in patients, related to breast risk. The modulation of nicotinamide adenine dinucleotide (NAD+) metabolism and the induction tumorigenic environment plays vital role progression. Among cell types, stem-like cells (CSCs) with self-renewal chemotherapy-resistance abilities could modulate tumor recurrence ability. In this study, we focused on visfatin's effect stemness-related properties using high-malignancy line MDA-MB-231 vitro vivo studies. Visfatin treatment significantly increased both sphere number diameter protein expression NANOG homeobox (NANOG), sex-determining region Y-box 2 (SOX2), octamer-binding transcription factor 4 (OCT4), as well SIRT1 levels. serum angiogenesis marker VEGF extracellular phosphoribosyl transferase (NAMPT, visfatin) were induced after visfatin treatment, increasing stemness environment, which reduced by inhibitor FK866. Our results demonstrate that visfatin-activated SIRT-SOX2 axis promotes triple-negative enriches microenvironment.
