Adipogenesis is intricately linked to the onset and progression of muscle aging; however, relevant biomarkers remain unclear. This study sought identify key genes associated with adipogenesis in context aging. Firstly, gene expression profiles from biopsies vastus lateralis both young elderly population were retrieved GEO database. After intersecting results differential analysis, weighted co-expression network sets adipogenesis-related genes, 29 expressed (ARDEGs) selected. Connectivity Map (cMAP) analysis identified tamsulosin, fraxidin, alaproclate as target compounds. In further, using three machine learning algorithms friends four hub ARDEGs, ESRRA, RXRG, GADD45A, CEBPB verified vivo aged mice muscles. Immune infiltration showed a strong link between several immune cells ARDEGs. all, these findings suggested that could serve related

Abstract Doxorubicin (Dox) is the standard treatment approach for osteosarcoma (OS), while acquired drug resistance seriously attenuates its efficiency. The present study aimed to investigate potential roles of metabolic reprogramming and related regulatory mechanism in Dox-resistant OS cells. results showed that ATP levels, lactate generation, glucose consumption oxygen rate were significantly increased cells compared with parental Furthermore, revealed expression estrogen-related receptor alpha (ERRα) was involved chemotherapy resistant cells, since targeted inhibition ERRα restored shifting profiles. Mechanistic analysis indicated mRNA stability, rather than transcription markedly chemoresistant Therefore, it hypothesized 3ʹ-untranslated region methylated by N 6 -methyladenine, which could further recruit insulin-like growth factor 2 binding protein 1 (IGF2BP1) suppress decay increase stability. IGF2BP1 knockdown downregulated reversed alteration Additionally, oncogenic effect IGF2BP1/ERRα axis on verified vitro vivo experiments. Clinical also levels associated clinical progression OS. Collectively, current suggested regulate contribute chemoresistance

Abstract Thiazolidinedione (TZD) is one of the privileged heterocyclic rings and has shown many biological applications in medicinal chemistry drug discovery. This review covers synthetic approaches TZD its derivatives, different techniques for affording desired regioselectivity stereoselectivity, that would enhance reaction conditions such as microwave, one‐pot, or ultrasound synthesis. It focuses on challenges glitazones transformation other heterocycles to TZD. Moreover, chemical behavior through substitution N3 position, modification C5 annealing complex systems, hybridization with pharmacologically attractive moieties are discussed. All reactions mentioned provided possible conditions, mechanisms, derivatives scope, yield clarified by construction potential agents. The also answers questions about rapid racemization glitazones, their toxicity, considering pan‐assay interference compounds (PAINS) not, influence saturation 5‐position activities. a comprehensive guide make informed decisions potentials.

Protein translation is an energy-intensive ribosome-driven process that reduced during nutrient scarcity to conserve cellular resources. During prolonged starvation, cells selectively translate specific proteins enhance their survival (adaptive translation); however, this poorly understood. Accordingly, we analyzed protein and mRNA transcription by multiple methods in vitro vivo investigate adaptive hepatic starvation. While acute starvation suppressed general, proteomic analysis showed induced of lysosome autolysosome proteins. Significantly, the expression orphan nuclear receptor, ESRRA (estrogen related alpha) increased served as a master regulator transcriptionally stimulating Rplp1 (ribosomal lateral stalk subunit P1) gene expression. Overexpression or siRNA knockdown Esrra led parallel changes expression, macroautophagy/autophagy translation, autophagy activity. Remarkably, have found had dual functions not only regulating but also controlling via ESRRA-RPLP1-lysosome-autophagy pathway
