The CRISPR Cas system, as a novel nucleic acid detection tool, is often hindered by cumbersome experimental procedures, complicated reagent transfer processes, and associated aerosol pollution risks. In this study, an integrated platform named "up down chip" was developed, which combined RT-RAA technology for amplification, DNase-dead Cas12a-modified magnetic beads specific recognition of target acid, HRP-TMB chromogenic reaction signal output in different chambers single microfluidic chip. were migrated up-and-down manner between through driving, achieving "sample-in, result-out" mode. By introducing homemade heating box temperature control during the using naked eye or smartphone APP color-based reading, no professional precise instruments required platform. Using platform, highly sensitive HIV-1 genome low 250 copies (CPs) per mL achieved within 100 min while maintaining good performance against common variants well excellent specificity anti-interference ability. addition, compared with qRT-PCR, it also exhibited accuracy 56 spiked plasma samples, indicating its promising potential clinical application.

Due to the sudden outbreak of COVID-19 at end 2019, rapid detection has become an urgent need for community clinics and hospitals. The development isothermal amplification technology nucleic acids in field molecular diagnostic point-of-care testing (POCT) gained a great deal attention recent years. Thanks intensive research on nicking enzymes, enzyme-combined promising platform detection. This is novel technique that uses enzymes improve ordinary amplification. Abstract Cerebral microbleeds are frequent incidental findings on brain MRI and have previously been shown to occur in Coronavirus Disease 2019 (COVID-19) cohorts of critically ill patients. We aimed determine the risk having medically indicated compare non-hospitalized COVID-19-infected patients with non-infected controls. In this retrospective case-control study, we included over 18 years age, an a susceptibility-weighted sequence, between 1 January July 2021. Cases were identified based positive reverse transcriptase polymerase chain reaction test for SARS-CoV-2 matched three non-exposed controls, sex, body mass index comorbidities. The number cerebral each scan was determined using artificial intelligence. 73 cases 219 COVID-19 associated significantly greater odds [odds ratio 2.66 (1.23–5.76, 95% confidence interval)], increasingly so when dementia hospitalized excluded. Our indicate that may be infections. This finding add pathophysiological considerations help explain previous COVID-19.
