Uncontrolled acute and persistent bleeding, as well with infection, is a great challenge because of the high mortality during treating patients injuries, complex surgery or bone marrow failure. Here, we develop an external form natural components which based on phosphorylated methacrylated gelatin (GelMA, G) cryogel (GP) loaded tannic acid (TA)-mixed copper ion (Cu2+) mesoporous bioactive glasses (MBG), named after GP@MBG-Cu-TA cryogel, to address goals reduce bleeding enhance antibacterial activity. Structurally, GP, MBG TA Cu2+ adheres GP via hydrogen bonding. In vitro, displays good biocompatibility, hemostatic antimicrobial capability. vivo studies, can effect in liver injury SD rats for aplastic anemia hemophilia A mice tail amputation bleeding. addition, accelerates skin wound repair bacterial contamination at site. sum, not only endowed dual function capability, but also stop objects either normal abnormal coagulation function. Thus, provides promising candidate dressing managing complications clinic.

A serious challenge of the chronic administration dexamethasone (DEX) is a delay in wound healing. Thus, this study aimed to investigate potential Tadalafil (TAD)-loaded proniosomal gel accelerate healing process skin wounds DEX-challenged rabbits. Skin were induced 48 rabbits 4 groups (n = 12 per group) and treated by sterile saline (control), TAD-loaded topically on wound, DEX-injected rabbits, DEX+TAD-loaded for weeks. The optical photography, transmission electron microscopy, vitro release profile, stability studies revealed successful preparation selected formula with good stability. DEX was associated uncontrolled oxido-inflammatory reactions, suppression immune response wounds, consequently failure process. gel-treated manifested marked enhancement rate closure than control (p < 0.05). successfully antagonized impacts dampening MDA production, enhancing total antioxidant capacity, coupled modulation inflammatory-related genes, inducible nitric oxide synthase, tumor necrosis factor-alpha, interleukin-1β, matrix metalloproteinase 9, during all stages This combination significant amplification response-related CD68 CD163 Moreover, histopathological, Masson’s Trichrome-stain, immune-histochemical indicated tissue recovery formation new blood vessels gel, as well-organized collagen fibers, upregulation transforming growth factor β1, vascular endothelial expression tissues Overall, topical application useful improving delayed linked therapy via regulating inflammatory/macrophage activation mediators enhanced angiogenesis, vascularity.
