The press coating approach has been employed in the current study; this system involves compressing directly of both core as well coat. procedure is same: first, we make tablet containing drug, diluent, binder, Crospovidone superdisintegrant for burst release after lag time, glidant, and lubricant an 8mm die. After that, get Eudragit S 100 polymer different ratios divide it into two equal parts, subsequently lower half inserted a cavity 10mm die followed by placed middle, other portion that comprises coat filled cavity. Powder's assessment parameters were judged to be within appropriate range. Overall, evaluation results press-coated tablets passed acceptable parameters. Optimized batch F7 shows 99.07% drug release. batches are further evaluated tests like FTIR, DSC, Stability study per standard ICH guidelines. From all result conclude concentration increases % also time. acts good time only at pH 7 means colon target was done hence successfully pulsatile delivery performed.

Theophylline is a drug commonly used to treat asthma due its anti-inflammatory and bronchodilatory properties. Testosterone (TES) has been suggested reduce the severity of symptoms. This condition affects boys more than girls in childhood, this ratio reverses at puberty. We reported that guinea pig tracheal tissue chronic exposure TES increases expression β2-adrenoreceptors enhances salbutamol-induced K+ currents (IK+). Herein, we investigated whether upregulation channels can enhance relaxation response methylxanthines, including theophylline. Chronic incubation tracheas with (40 nM, 48 h) enhanced induced by caffeine, isobutylmethylxanthine, theophylline, an effect was abolished tetraethylammonium. In myocytes, increased theophylline-induced IK+; flutamide reversed effect. The increase IK+ blocked 4-aminopyridine ~82%, whereas iberiotoxin reduced ~17%. Immunofluorescence studies showed KV1.2 KV1.5 airway smooth muscle (ASM). conclusion, ASM promotes theophylline response. Therefore, gender should be considered when prescribing as teenage males are likely respond better females.
