Atherosclerosis (AS) is the pathology of atherosclerotic cardiovascular diseases (ASCVD), characterized by persistent chronic inflammation in vessel wall, which monocytes/macrophages play a key role. It has been reported that innate immune system cells can assume proinflammatory state after short stimulation with endogenous atherogenic stimuli. The pathogenesis AS be influenced this hyperactivation system, termed trained immunity. Trained immunity also implicated as pathological mechanism, leading to AS. mediated via epigenetic and metabolic reprogramming occurs mature their bone marrow progenitors. Abstract Osteoporosis (OP) is a common metabolic bone disease. Excessive osteoclastic activity significantly contributes to the development of OP. Icariin (ICA) flavonol glycoside derived from herbal plants and possesses curative effects on postmenopausal OP fracture. This study aimed investigate ICA osteoclast differentiation induced by receptor activator nuclear factor kappa B (RANK) ligand (RANKL) involvement estrogen α (ER ) RANK signaling cascade in this process. RANKL was used induce RAW264.7 cells into osteoclasts. Small interfering RNA technique knockdown ER cells. Cell counting kit-8 assay performed determine cytotoxicity ICA. The number tartrate-resistant acid phosphatase (TRAP)-positive quantified TRAP staining. osteoclasts, while abolished pro-osteoporotic effect RANKL. Moreover, RANKL-induced osteoclastogenesis. Further exploration revealed that inhibited phosphorylation c -Src osteoclasts via regulating , inactivation reversed knockdown-promoted Lastly, activation mitogen-activated protein kinase pathway downregulated expressions target osteoclastogenic proteins RANKL-treated RAW 264.7 cells, almost completely diminished / -Src/RANK signaling. These findings elucidated novel mechanism which exerts an anti-osteoporotic effect.
